摘  要:目的研究模拟的锰超氧化物歧化酶(mimic manganese superoxide dismutase,Mn SODm)对急性单核细胞白血病细胞(U937细胞)凋亡和增殖的影响及部分机制。方法以不同剂量的Mn SODm(1、2、3和4mg·L-1)处理U937细胞后,用MTT法检测U937细胞的增殖率,用Annexin V-PE/7-AAD双染法检测U937细胞的凋亡率。用蛋白印迹法测定cleaved-caspase-8、cleaved-caspase-9和cleaved-caspase-3蛋白的表达。结果 Mn SODm处理组U937细胞的增殖率明显低于对照组(P<0.05);Mn SODm能够有效地抑制U937细胞的增殖,这种抑制作用呈现明显的浓度依赖性及时间依赖性(P<0.05)。Mn SODm处理组U937细胞的凋亡率明显高于对照组(P<0.05);Mn SODm的凋亡诱导作用具有明显的浓度依赖性(P<0.05)。蛋白印迹法结果显示cleaved-caspase-8、cleaved-caspase-9和cleaved-caspase-3蛋白的表达亦呈现出浓度依赖性上升的趋势。结论 Mn SODm能够有效地抑制U937细胞的增殖,促进U937细胞的凋亡,其作用具有浓度依赖性及时间依赖性。Objective To explore effects and mechanism of Mimic manganese superoxide dismutase in inhibiting proliferation and inducing apoptosis in human acute monocytic leukemia U937 cells in vitro. Methods MTT assay and AnnexinV - PE/7 - ADD staining assay was adopted to observe the proliferation inhibition and apoptosis induction effects of MnSODm. Apoptosis related proteins expression include caspase - 8, caspase - 9 and caspase -3 were detected by western blotting assay. Results MTY assay shows MnSODm markedly re- duced the proliferation of U937 cells versus control group (P 〈 0.05 ). MnSODm reduced the proliferation of U937 cells in a time and dose - dependent manner (P 〈 0.05 ). MnSODm markedly highlight apoptosis rate of U937 cells in a dose- dependent manner(P 〈0.05 ). The western blotting results also show that the expression of cleaved - caspase - 8, cleaved - caspase - 9 and cleaved - caspase - 3 were enhanced in a dose - dependent manner. Conclusion MnSODm could inhibit acute monocyte leukemia cells in a time and dose - dependent manner via decreasing proliferation and induction apoptosis.
