摘  要:目的探讨黄芪甲苷对甲基苯丙胺(MA)依赖大鼠脑组织bcl-2及caspase-3蛋白表达的影响。方法选用健康Wistar雄性大鼠60只,随机分为空白对照组、MA依赖模型组、黄芪甲苷(10 mg/kg)+MA干预组和黄芪甲苷(20 mg/kg)+MA干预组,每组15只。空白对照组腹腔注射生理盐水;MA依赖模型组及药物干预组按剂量逐日递增法腹腔注射给药,建立MA依赖大鼠实验动物模型;同时灌胃给药黄芪甲苷干预治疗。应用免疫组化法检测大鼠脑组织bcl-2及caspase-3蛋白的表达;应用RT-PCR和琼脂糖凝胶电泳方法测定bcl-2、caspase-3 mRNA的表达。结果与空白对照组比较,MA依赖模型组脑组织的caspase-3表达增加(P<0.01),bcl-2在应激状态下也有明显表达;大剂量黄芪甲苷处理组与MA依赖模型组比较bcl-2表达显著增加(P<0.01),caspase-3蛋白表达明显减少(P<0.01);bcl-2 mRNA的表达显著增加,caspase-3 mRNA的表达明显减少(均P<0.01)。结论黄芪甲苷可明显增加MA依赖大鼠脑组织bcl-2蛋白表达,降低caspase-3蛋白表达作用,并上调bcl-2 mRNA表达和下调caspase-3 mRNA表达的作用。Objective To investigate the effect of astragaloside Ⅳ on bcl-2 and caspase-3 expression in methamphetamine dependent rat. Methods A total of 60 healthy Wistar rats were randomly divided into control group,MA dependente model group,astragaloside Ⅳ( 10 mg / kg) + MA intervention group and astragaloside Ⅳ( 20 mg / kg) + MA intervention group( n = 10 each group). Control group were injected with 0. 9% NS; methamphetamine dependent rat model was established in MA dependent group and astragaloside Ⅳ + MA group according to dailydose escalation method. bcl-2 and caspase-3 protein expression changes were detected by immunohistochemistry method; RT-PCR and agarose gelelectrophoresis was used to detect the expression of bcl-2 and caspase-3 mRNA. Results Compared with the control group,MA dependent of brain tissue bcl-2 and caspase-3 expression was increased( P〈0. 01); Compared with MA dependent model group,astragaloside Ⅳ( 20 mg / kg) treatment group,bcl-2 expression a was significantly increased( P〈0. 01),caspase-3protein expression was significantly reduced( P〈0. 01). Conclusion Astragaloside Ⅳ can up-regulate bcl-2 protein expression in the MA dependent model group,while also reduce caspase-3 protein expression in the role of the brain tissue.
