摘  要:Let-7是最早发现的miRNA之一,参与动物多个器官发育的调控过程,也在多种癌症发生中发挥抑癌作用。本研究采用Rt-qPCR(PCR)方法首先检测金华猪不同发育时期肝脏组织中let-7家族成员之一的miR-let-7i的表达变化,同时比较金华猪和大约克猪心、肝、脾、肺、肾、胰脏、小肠和皮脂中miR-let-7i的表达差异。结果表明:miR-let-7i在金华猪肝脏发育过程中表达总体趋势为胚胎期低于生长阶段,幼年期低于成年期;比较不同猪种间组织表达差异发现,在胚胎80d时,miR-let-7i在小肠中的表达量最高,在心、肝、脾、肺、小肠、皮脂中,金华猪的相对表达量极显著高于大约克(P<0.01),而在肾和胰脏中的表达却极显著低于大约克(P<0.01)。在出生后90日龄时,miR-let-7i在金华猪的心、肺和小肠中表达较高,在约克猪的胰、心和脾脏中表达较高;金华猪的肝、脾、胰脏中miR-let-7i的表达量极显著低于大约克(P<0.01),而在肺、肾、小肠、皮脂中极显著高于大约克(P<0.01)。本研究为探究miR-let-7i在动物发育过程的功能和分子机制奠定了基础。Let-7 is one of the earliest discovered miRNAs, which participates in the regulation of many organ developmental processes and play a tumor suppressor function in some cancer formation. In the present study, the expression of miR-let-Ti, a member of the let-7 family, was detected in Jinhua pig liver at various developmental stages by Rt-qPCR (Real-time quantitative PCR), and miR-let-7i differential expression pattern in various tissues was analyzed between Jinhua and Yarkshire. The results showed that the general expression of let-7i in embryonic period was lower than that in the growth stage, the juvenile stage is lower than the adulthood. At embryo d80 , miR- let-7i was abundantly expressed in small intestine; compared to Yorkshire, miR-let-7i was higher expressed in heart, liver, spleen, lung, small intestine and sebum of Jinhua pig (P〈0.01), respectively, while in kidney and pancreas the expression was significantly lower (P〈0.01). At d 90 after birth, the expression of miR-let-7i in liver, spleen and pancreas of Jinhua was lower than that of Yarkshire; lung, kidney, small intestine and sebum, the expression of Jin hua was lower than that of Yorkshire (P〈0.01). This study could provide the base for exploring the roles of miR-let-7i in animal development.
