摘  要:目的观察舒洛地特软胶囊对糖尿病(DM)大血管病变大鼠血管内皮生长因子(VEGF)水平的影响,并了解其对糖尿病大血管病的作用。方法建立糖尿病大血管病变SD大鼠模型,干预组给予舒洛地特混悬液灌胃;8周末处死大鼠,取血测生化指标,酶联免疫吸附法测血浆VEGF水平,留取胸主动脉用免疫组织化学法测定组织VEGF表达。结果实验性DM大血管病变大鼠模型的生化代谢指标、病理结果符合糖尿病大血管病变临床疾病表现。DM组血浆、组织VEGF测定结果较正常对照组升高;DM干预组血浆、组织VEGF水平较DM对照组下降;DM干预组血管病变较DM对照组减轻。结论高血糖可能诱导VEGF表达增加,从而参与DM大血管病变的发生、发展;舒洛地特软胶囊可能通过降低VEGF表达发挥DM大血管病变的保护作用。【Objective】 To observe the sulodexide soft capsules on vascular endothelial growth factor(VEGF) level in rats with diabetic macroangiopathy, and to observe its possible effects on diabetic vascular disease. 【Methods】 Diabetic macroangiopathy model was established in SD rat. Sulodexide was given to the rats of the intervention group. At the end of 8th week, all the rats were killed. Blood was obtained for biochemical tests. Plasma VEGF level was measured by ELISA. VEGF expression in the tissue of thoracic aorta was determined by immunohistochemical method. 【Results】 The biochemical indicators and pathological results in the experimental diabetic macroangiopathy rat model were consistent with the clinical manifestations of the disease. The plasma and tissue VEGF levels in the diabetes mellitus(DM) group increased compared to those in the normal control group. VEGF expression in the DM intervention group declined markedly compared to that in the DM control group. The angiopathy was alleviated in the DM intervention group compared with that in the DM control group. 【Conclusions】 The hyperglycemia may induce increased VEGF expression, thereby participate in the occurrence and development of diabetic macroangiopathy. Superoxide soft capsule may play a protective effect on macroangiopathy in DM by reducing VEGF expression.
