黄连素对大鼠脊髓损伤后细胞自噬与凋亡的影响

摘　　要：目的 观察黄连素对大鼠脊髓损伤后细胞自噬与凋亡的影响.方法 128只雌性SD大鼠随机分为3组.A组：假手术组（n=32）,B组：Allen＇s法脊髓损伤组（n=48）,C组：T10脊髓损伤＋黄连素干预组（n=48）.采用大鼠运动功能Basso-Beattie-Bresnahan（BBB）评分分别评估各组大鼠在模型建立后6h、1、3、5d的运动功能,Western blot分别检测各组上述时间节点脊髓损伤区域微管相关蛋白1轻链3（LC3）-Ⅱ及半胱氨酰天冬氨酸特异性蛋白酶（Caspase）-3的表达.结果 C组BBB评分在术后3、5d分别较B组升高[（2.44±0.92）比（1.33±0.76）分、（5.87±2.02）比（3.66±1.48）分,P ＜0.05];C组LC3-Ⅱ相对表达量在术后6h、1、3、5d分别较C、A组升高（0.63±0.19比0.41 ±0.12、0.25 ±0.03;0.71 ±0.21比0.45 ±0.18、0.28 ±0.16;0.75 ±0.17比0.57±0.21、0.26 ±0.12;0.69 ±0.13比0.52±0.23、0.25±0.14,P＜0.05）;C组Caspase-3相对表达量在术后6h、1、3、5d分别较C、A组升高（0.81 ±0.20比0.61 ±0.17、0.51 ±0.13;0.84 ±0.28比0.62±0.14、0.48 ±0.18;0.91 ±0.31比0.68±0.23、0.52 ±0.15;0.87 ±0.25比0.61 ±0.21、0.45 ±0.16,P ＜0.05）.结论 黄连素可以上调大鼠脊髓损伤组织的自噬活性,并通过低表达Caspase-3减少细胞凋亡,从而促进神经功能恢复.Objective To observe the effects of berberine on autophagy and apoptosis after spinal cord injury （SCI） in rats.Methods 128 female SD rats were randomly divided into three groups：group A,sham operation group （n =32）;group B,Allen＇s SCI group （n =48）;and group C,T10 SCI ＋ berberine-treated group （n =48）.The motor function of rats with score [Basso-Beattie-Bresnahan （BBB） score] was used to evaluate motor function of SCI rats at 6th h,1st day,3rd day,and 5th day.Western blotting was used to detect the expression of microtubule-associated protein 1 light chain 3 （LC3）-Ⅱ and Caspase-3 in SCI.Results BBB scores in group C on the postoperative day 3 and 5 were significantly increased as compared with those in group B [（2.44 ± 0.92） vs.（1.33 ± 0.76）;and （5.87±2.02） vs.（3.66±1.48）;P＜0.05].The relative expression of LC3-Ⅱ in group C on the postoperative 6th h,1st day,3re day,and 5th day was significantl increased as compared with group C,group A [（0.63 ±0.19） vs.（0.41 ±0.12）,（0.25 ±0.03）;（0.71 ±0.21） vs.（0.45 ±0.18）,（0.28±0.16）;（0.75±0.17） vs.（0.57±0.21）,（0.26±0.12）;（0.69±0.13） vs.（0.52±0.23）,（0.25 ±0.14）,P ＜0.05].The relative expression of Caspase-3 in group C on the postoperative 6th h,1st day,3rd day,and 5th day was significantly increased as compared with group C,group A [（0.81 ± 0.20） vs.（0.61±0.17）,（0.51 ±0.13）;（0.84±0.28） vs.（0.62±0.14）,（0.48±0.18）;（0.91 ±0.31） vs.（0.68 ±0.23）,（0.52±0.15）;（0.87 ±0.25） vs.（0.61 ±0.21）,（0.45 ± 0.16）,P ＜ 0.05].Conclusion Berberine can enhance the autophagic activity in rats with SCI,and promote the recovery of neurological function through reducing apoptosis via down-regulating the expression of Caspase-3.
