摘  要:为了探讨栽培年限对亳芍中芍药总苷类成分含量的影响,为亳芍规范化栽培、质量评价提供依据,本文建立同时测定亳芍中芍药苷和芍药内酯苷含量的高效液相色谱检测方法,采用Lanbo Service 4000高效液相色谱仪、Waters Symmetry C18色谱柱(4.6 mm×150 mm,5μm);以乙腈:0.1%磷酸溶液梯度洗脱;流速1.0 m L/min;检测波长230 nm;柱温30℃。结果表明,芍药苷和芍药内酯苷色谱峰分离度较好,分别在0.1250~2.5000μg和0.0225~0.4500μg呈良好的线性关系,相关系数分别为0.9960和0.9994,加样回收率(n=6)分别为100.56%、101.30%。该方法用于安徽亳州地区人工栽培1至7年的亳芍中芍药苷及芍药内酯苷的测定,发现不同年栽培年限亳芍中均含有芍药苷和芍药内酯苷,二者的含量存随年份变化表现出一定差异,其中以3年株龄亳芍中芍药苷含量最高,以2年株龄亳芍中芍药内酯苷含量最多。研究结果对亳芍临床用药有一定参考价值。In order to explore the accumulation of glucosides in Bozhou Peony Paeonia lactiflora with different cultivation ages and to provide references for the standardized cultivation and quality evaluation of Bozhou Peony,a HPLC-DAD method was developed and applied for the simultaneous determination of paeoniflorin and albiflorin in P. lactiflora. A Lanbo Service 4000 system was used with Waters Symmetry C18column( 4. 6mm × 150 mm,5μm) at 30 ℃ with a gradient elution of acetonitrile and 0. 1% phosphonic acid. The flow rate was 1. 0 m L / min and the detection wavelength was230 nm. The calibration curves of paeoniflorin and albiflorin were linear over the ranges of 0. 1250-2. 5000 μg and0. 0225-0. 4500 μg. The average recoveries( n = 6) were 100. 56% and 101. 30%,respectively. The developed method was successfully applied for the analysis of Bozhou Peony P. lactiflora of different plant ages from its Geo-authentic habitats-Bozhou region,Anhui province,China. The results also indicated that remarkable variation of the contents of paeoniflorin and albiflorin were existed in Bozhou Peony with different plant ages. In our experimental conditions,the highest contents of paeoniflorin and albiflorin were the Bozhou Peony cultivated for 3 years and 2 years,respectively. Our research results provided some references for the clinical medication of Bozhou Peony.
