摘 要:目的:利用高脂饲料建立非酒精性脂肪肝动物模型,为非酒精性脂肪肝的中药治疗效果提供基础。方法:将70只清洁级SD大鼠随机分为2组,即正常组20只、模型组50只,正常组随机分为5组,每组4只;模型组随机分为5组,每组10只。正常组给予普通饲料喂养,模型组给予高脂饲料喂养。于第四周、第六周、第八周、第十周、第十二周分别处死1组正常组和1组模型组。观察大鼠肝脏大体观,称量肝脏湿重,计算大鼠肝体比,并将肝脏进行石蜡切片HE染色和冰冻切片苏丹三染色。结果:从第四周末开始,模型组大鼠肝湿重增高,与正常组大鼠肝湿重比较差异具有统计学意义,并在第六周末、第十周末、第十二周末出现显著差异。从第六周末开始模型组肝指数增高与正常组肝指数比较差异具有统计学意义。肝脏镜下观从第四周开始出现轻度脂肪肝,并随着造模时间的延长,不断加重。结论:利用常规高脂饲料(88%普通饲料中加入10%猪油和2%胆固醇)造非酒精性脂肪肝动物模型,于造模后第四周即出现非酒精性脂肪肝,并随着时间延长不断加重。Objective The high fat diet to establish the animal model of nonalcoholic fatty liver and thorough research on non alco- holic fatty liver of traditional Chinese medicine treatment provide the basis. Methods 70 male SD rats were randomly divided into two groups, namely the normal group (n =20), model group (n =50) and normal group were divided to five groups, 4 rats in each group; model group was divided for 5 groups with 10 rats in each group. The normal group was given normal diet, model group was given conventional high fat diet. In the fourth week, sixth weeks, eighth weeks, tenth weeks, Twelfth weeks respectively 1 normal grup and 1 model grup nre exeuted. Observation of rat liver grossly, weighing liver wet weight calculated rat liver body ratio, and liver were paraffin section and he staining and frozen sections with Sudan Ill staining. Results From the beginning of the fourth week, rats in the model group liver wet weight increased, normal rats liver wet and heavy difference, and there was significant difference in the sixth week, the tenth week and the twelfth week. From the sixth weekend model group liver index increased significantly difference with normal liver index contrast group. The microscopic appearance of liver from four appeared mild fatty liver, and with the extension of model making, increasing. Conclusion The conventional high fat diet (88% ordinary feed adding 10% lard oil and 2% cholesterol) made the animal model of nonalcoholic fatty liver. After modeling the around the nonalcoholic fatty liver, and with the prolongation of time increasing.
