摘  要:目的:建立HPLC法测定人血浆中吗啡浓度的方法。方法:采用高氯酸为蛋白沉淀剂,以及采用色谱枉为、vaters Spherisorb C18 ODS柱,流动相为0.05mol/L磷酸二氢钾溶液.0.0025mol/L庚烷磺酸钠水溶液-乙晴(5:5:2),柱温为30℃,流速为0.5mL/min,检测波长为220nm,进样量为20μL。结果:血浆中吗啡浓度是在1.0~100μg/mL范围内其线性关系良好(r=0.9994);最低检出限为0-3μg/mL;低、中、高3种吗啡浓度的回收率分别为98.70%,113.71%和96.51%:日内RSD分别为3.39%,4.96%和2.27%;日间RSD分别为7.64%,5.74%和3.18%。结论:该方法操作简便、快速,可适用于人血浆中吗啡浓度的测定。Objective: To establish a method to determine rapidly morphine in human palsma by HPLC. Methods: The separation was performed on Waters Spherisorb C18 column with 0.05 mol/L potassium dihydrogen phosphate-0.002 5 mol/L sodium heptanesulfonate-acetonitrile(5 : 5: 2) as mobile phase using perchloric acid as protein precipitant. The column temperature was 30 ℃, the flow rate was 0.5 mL/min, the detection wavelength was 220 nm, and the sam- ple volume was 20 μL. Results: The calibration curve of morphine was linear within the range of 1.0-100 μg/mL with good linearity(t=0.999 3 ). The lowest detection limit was 0.3 μg/mL. The recovery was 98.70%, 113.71% and 96.51% with intra-day RSD of 3.39%, 4,96% and 2.27% and inter-day RSD of 7.64%, 5.74% and 3.18% for low, medium and high concentration, respectively. Conclusion: This method is simple, rapid and stable for the determination of morphine concentration in human plasma.
