光生物反应器培养聚球藻7002产嗜铁素的优化方法

摘  要:为了缩短聚球藻7002的培养周期、提高嗜铁素的产量,使用10L封闭式光生物反应器在铁限制条件下光自养培养聚球藻7002,研究光照强度、温度、CO2体积分数、通气量4个环境因素,以及接种比对嗜铁素分泌的影响,确定产生嗜铁素最优条件为:光照强度140μmol/(m2·s)、温度30℃、CO2体积分数0.5%、通气量1.5L/min、接种比1∶25。优化后嗜铁素含量终值可达(92.35±1.36)%,比摇瓶培养方式提高了49.29%;3d即可采收,比摇瓶培养提前了15d。In order to shorten culture period of Synechococcus sp .PCC 7002,increase the production of siderophores,an 10 L photobioreator was used for autophototrophic cultivation of Synechococcus sp .PCC 7002 uncler iron limited condition.Then the effects of inoculating population ratio and four environmental factors including light intensity,temperature,the concentration of CO2 (V/V)and aeration rate on siderophores production were studied.The optimum parameters of siderophores secretion are as follows:light intensity is 140 μmol/(m2 ·s),temperature is 30 ℃,the concentration of CO2 (V/V)is 0.5%,aeration rate is 1.5 L/min and the inoculating population ratio is 1:25.The final siderophore unit can be (92.35±1.36)% which is 49.29% higher than that when cultivated in shaking flask,and the harvest time can be the 3rd day since inoculation which is 15 days earlier than that when cultivated in shaking flask.
