摘 要:目的:建立高效液相色谱法(high performance liquid chromatography,HPLC)测定奥扎格雷钠脂质体包封率的检测技术。方法采用HPLC,以C18色谱柱分离,以0.05 mol/L磷酸二氢钾溶液( pH=7.0)-甲醇-乙腈(80∶15∶5)为流动相,检测波长为274 nm测定。结果奥扎格雷在10~100μg/ml浓度范围内线性关系良好,r=0.9997。日内、日间RSD均小于0.5%,方法回收率为(100.2±0.42)%,样品的平均包封率为(94.89±0.44)%。结论该测定方法简单,准确度高,灵敏度好,适合奥扎格雷钠脂质体的包封率测定。Objective To establish a high performance liquid chromatography ( HPLC) method for the encapsulation efficien-cy of ozagrel hydrochloride liposome.Methods The HPLC was used for determination.A C18 column was used with the mobile phase of 0.05 mol/L potassium dihydrogen phosphate solution (pH=7.0) -acetonitrile-methanol (80∶15∶5) at the detection wavelength of 274 nm.Results The calibration curve was linear in the concentration range of 10-100 μg/ml, r=0.9997.The inter-and intra-day RSD was less than 0.5 %.The method recoveries were (100.2 ±0.42) %.The average entrapment efficiency of tartrate in-doquinoline liposome was (94.89 ±0.44)%.Conclusions This method is simple,accurate and can be used to determine the encap-sulation efficiency of ozagrel hydroehloride in ozagrel hydrochloride liposome.
