摘 要:建立了高效液相色谱同时测定苦楝子中的三种有效成分(香草酸、香兰素、阿魏酸)的方法。苦楝子样品用甲醇超声提取,提取液过滤并用甲醇稀释后分析。采用C18反相色谱柱(250×4.6nm,3.5μm),流动相为乙腈和0.1%醋酸水溶液,梯度洗脱。结果显示:三种有效成分的检出限(LOD)分别为2.5μg/m L、0.82μg/m L和1.3μg/m L,定量限(LOQ)分别为8.4μg/m L、2.7μg/m L和3.4μg/m L,加标回收率为94.9%~104.5%,相对标准偏差小于3.4%,苦楝子样品中的香草酸、香兰素、阿魏酸的平均含量分别是67.98 mg/kg、96.14 mg/kg、36.61 mg/kg(n=6),相对标准偏差分别为1.8%、1.1%和0.7%。该方法重复性好、简单、准确,可用于同时测定苦楝子中的香草酸、香兰素、阿魏酸的含量。A high performance liquid chromatography method for analysis of three effective components in Melia azedarach L.was developed. The components were extracted from the samples in methanol under ultrasonic irradiation and diluted with methanol after filtration. The analysis was performed on a C18 column(250×4.6nm,3.5μm) using a gradient elution program with the mobile phase of 0.1% acetic acid solution and acetonitrile.The results show that the limits of detection(LODs) for three effective components are 2.5 μg/m L,0.82 μg/m L and 1.3 μg/m L,and the limits of quantification(LOQs) are 8.4 μg/m L,2.7 μg/m L and 3.4 μg/m L.The average recoveries are in the range of 94.9%~104.5% for three effective components, the relative standard deviations are below 3.4%. The contents of three effective components in Melia azedarach L.are 68.0 mg/kg,96.1 mg/kg and 36.6 mg/kg(n=6),respectively, recoveries are 1.8%,1.1% and 0.7%. This method is reliable, sensitive and reproducible, and it can be used for determination of three effective components(Vanillic Acid,Vanilline and Ferulic Acid) in Melia azedarach L.
