摘  要:采用苯酚-硫酸法测定山银花(Lonicera hypoglauca)多糖的含量,并对提取工艺进行优化。在单因素试验的基础上,采用L9(34)正交试验,以山银花多糖提取率为指标,探讨了提取次数、提取温度、提取时间、料液比对山银花多糖提取率的影响。结果表明,最佳提取工艺参数为提取4次、提取温度80℃、提取时间150 min、料液比1∶20(g∶m L)。在此条件下,山银花中多糖平均提取率为15.39%,加样回收率为97.70%,该方法提取山银花多糖操作简便、准确度高、重现性好,可作为山银花多糖提取和含量测定的方法。In this study, the content of polysaccharide in Lonicera hypoglauca was determined by phenol sulfuric acid method and the parameters of extraction process was optimized. Based on the single factor experiment, using L9(34) orthogonal exper- iment, with the content of polysaccharide in Lonicera hypoglauca as the indicator, effect of extraction times, extraction tem- perature, extraction time, solid-liquid ratio on the extraction rate was studied. The results showed that the optimum extraction process parameters were as followed: extracting 4 times, extraction temperature 80℃, extraction time 120 min, ratio of solid to liquid 1:20 (g/mL). Under such conditions, the average extraction rat of polysaccharide in Lonicera hypoglauca was 15.39%, the average recovery rate was 97.70%. The method for extraction of polysaccharide in Lonicera hypoglauca had the advantages of simple operation, high accuracy, good reproducibility. Therefore it can be used in the extraction and determi- nation of the content of the polysaccharide.
