摘  要:目的:研究复方中药益气养血补肾方对长期大强度运动大鼠海马JAK/STAT信号转导通路的影响,进一步探讨该中药的神经保护作用及机制。方法:30只雄性SD大鼠随机分为对照组、大强度运动组和中药组,每组10只。其中,大强度运动组和中药组进行递增负荷跑台训练,时间为7周。用TUNEL法检测大鼠海马细胞凋亡情况,用Western-blotting法测定海马磷酸化JAK1、STAT1及Casepase-3的蛋白表达水平。结果:与对照组相比,大强度运动组细胞凋亡水平和p-JAK1、p-STAT1和Casepase-3的蛋白表达水平均显著升高(P<0.01);中药组细胞凋亡水平和p-JAK1、p-STAT1和Casepase-3的蛋白表达水平则明显低于大强度运动组(P<0.05,P<0.01)。结论:长期大强度运动可能激活JAK/STAT信号转导通路,发挥促凋亡作用,参与脑缺血损伤后的病理生理过程;复方中药益气养血补肾方可能通过抑制pJAK1、p-STAT1和Casepase-3的表达发挥脑缺血后的抗凋亡作用,从而达到神经保护作用。Objective: To study the effects of compound Chinese medicine on the signal pathway of JAK / STAT in rat hippocampus with long- term high intensity exercise. Methods: Male SD rats were randomly divided into three groups: control group( n = 10),high intensity exercise group( n = 10)and Chinese medicine group( n = 10). The exercise project was applied by 7 weeks treadmill at progressive increase exercise intensity. The level of apoptosis in rat hippocampus was detected by the methods of TUNEL. The level of protein expression of p- JAK1,p- STAT1 and Casepase- 3 were determined by the methods of Western blotting. Results: Compared with the control group,the level of apoptosis and protein expression of p- JAK1,p- STAT1 and Casepase- 3 in high intensity exercise group were obviously increased. The level of apoptosis and protein expression of p- JAK1,p-STAT1 and Casepase- 3 in Chinese medicine group were significantly decreased compared with those in high intensity exercise group. Conclusions: The JAK / STAT signal pathway which may be activated by long- term high intensity exercise participates in pathophysiologic process of cerebral ischemia by promoting apoptosis possibly. The compound Chinese medicine can reduce the expression of p- JAK1,p- STAT1 and Casepase- 3 and play an important role in antiapoptosis so as to protect the brain from ischemia injury probably.
