摘 要:目的研究耐力运动和脂联素受体激动剂(AdipoRon)单独或联合干预对高脂饮食小鼠脂肪组织炎症的影响。方法 50只雄性C57BL/6小鼠随机分为普通饮食对照组(NC)、高脂饮食对照组(HC)、高脂饮食运动组(HE)、高脂饮食+AdipoRon组(HA)和高脂饮食+运动+AdipoRon组(HEA),每组10只。HE和HEA组进行6周中等强度耐力跑台训练;HA和HEA组按50 mg/kg剂量隔天皮下注射25%AdipoRon盐溶液。检测小鼠血清糖、脂代谢参数和炎症因子;腹腔注射葡萄糖耐量实验(IPGTT)检测小鼠糖耐量;RT-PCR法检测附睾脂肪基因表达。结果与NC组相比,HC组小鼠血清TG、TC、FFA、FBG和促炎因子TNF-α、IL-6浓度显著升高(P<0.05);TNF-α、IL-6、F4/80、CD11c、HIF-α、PDK1、GLUT1mRNA表达显著升高(P<0.05),IL-10 mRNA表达显著下降(P<0.05),糖耐量明显受损。HE和HA组小鼠除FBG、IL-10外,各项指标均显著高于NC组(P<0.05);与HC组相比,HE和HA组小鼠血清TG、TC、FFA、FBG、TNF-α、IL-6浓度和TNF-α、IL-6、F4/80、CD11c mRNA表达显著下降(P<0.05),IL-10表达显著升高(P<0.05),HIF-α、PDK1、GLUT1 mRNA表达下降无显著性差异;HEA组血清TG、FFA、TNF-α浓度和TNF-α、IL-6、F4/80、HIF-α、PDK1、GLUT1 mRNA表达较HE和HA组具有显著性差异(P<0.05),HE、HA和HEA组糖耐量得到显著改善。结论有氧运动和AdipoRon可能通过改善脂肪组织缺氧来降低高脂饮食小鼠脂肪组织巨噬细胞数量和M1态激活,降低脂肪组织炎症,同时改善葡萄糖耐受受损和脂代谢紊乱,并且联合干预效果优于单一干预效果。Objective The effects of endurance exercise and adiponectin receptor agonist--AdipoRon on adipose tissue in- flammation of high fat diet feeding mice were investigated. Methods 50 male C57BL/6 mice were randomly divided into nor- mal diet control(NC) group, high fat diet control(HC) group, high fat diet + exercise(HE) group, high fat diet + Adi- poRon(HA) group and high fat diet + exercise + AdipoRon(HEA) group with 10 mice in each group. HE and HEA group were subjected to an 6 - week moderate endurance treadmill running program while HA and HEA group received subcutane- ous injection of 25% AdipoRon salt solution at a dose of 50 mg/kg body weight every other day. Serum glucose, lipid meta- bolic indicators and cytokines were determined. IPGTr test was performed to evaluate the glucose tolerance and RT - PCR was used to evaluate gene expression of epididymis fat. Results Compared with NC group, serum TG, TC, FFA, FBG and pro - inflammatory cytokines TNF -α, IL - 6 concentration was significantly elevated( P 〈 0.05 ) ; TNF - α, IL - 6, F4/80, CD11c, HIF-α,PDK1 and GLUT1 mRNA expression was increased markedly (P 〈 0. 05 ) while IL- 10 was reduced sig- nificantly( P 〈 O. 05 ) ; ,Glucose tolerance is obviously impaired . Besides FBG and IL - 10, other indicators of HE and HA group were significantly higher than NC group. Compared with HC group, however, serum TG, TC, FFA, FBG, TNF -α, IL- 6 concentration and TNF-α,IL- 6, F4/80, CDllc , HIF-α, PDK1, GLUT1 mRN A expression were reduced signif- icantly ( P 〈 0.05 ) , while IL - 10 mRNA was elevated significantly in HE, HC. And serum TG, FFA, TNF -α and TNF - ,IL - 6, F4/80, HIF - α, PDK1 ,GLUT1 mRNA expression were further reduced in HEA group ( P 〈 O. 05 ). In addi-tion, glucose tolerance of HE, HA and HEA group is significantly improved.
