摘  要:由于血药浓度可反映药物在体内(靶器官)的状况,准确测定血药浓度在设计和优化临床给药方案和药动学研究方面发挥着越来越重要的指导作用。LC-MS/MS因其高灵敏度、高准确性、高专属性的特点在药学研究领域尤其是定量测定血药浓度方面被广泛应用。然而,由于血液成分复杂,基质效应成为影响血药浓度测定结果的可靠性、准确度及精密度的主要因素。本文通过查阅国内外近几年的文献,对血药浓度测定中基质效应的可能机制、评价方法、验证方法及解决方案进行了综述。The blood concentration may reflect the situation of the drug in body( target organ),so that accurate determination of drug concentration in blood plays an increasingly important role in optimizing clinical administration and pharmacokinetics. LC-MS / MS is widely used in pharmacological research fields,especially in the quantization of blood concentration because of its high sensitivity,high accurate,and high specificity. However,due to the complexity of blood components,matrix effect becomes a major factor affecting the reliability,accuracy and precision of LC-MS / MS determination. Therefore,in this paper,authors reviewed the possible mechanisms,assessment method,validation method and strategies for elimination of matrix effect via citing the articles published in recent years.
