摘 要:采用PCR方法扩增了22个传染性喉气管炎病毒(ILTV)田间分离株的ICP4基因片段,并与GenBank收录的部分鸡胚源(CEO)弱毒疫苗毒株、细胞培养源(TCO)弱毒疫苗毒株和国内分离强毒株进行了序列比较。结果显示:所有田间分离株和国内近期分离的LJS09毒株所测定,C刚基因片段均为676bp;与国内早期分离的王岗(wG)株7LTCO疫苗株相比,田间分离株和CEO疫苗株在ICP4基因的272~283nt部位均存在12个碱基的缺失;遗传进化树显示,田间分离株与CEO疫苗株的距离较近,处于同一个大分支上,而与TCO疫苗株及WG株的距离相对较远,处于不同分支上。上述结果表明,我国ILTV田间分离株的遗传特性与CEO疫苗株接近。The ICP4 gene fragments from 22 field isolates of infectious laryngotracheitis virus (ILTV)were amplified by PCR method,and then the sequences were aligned with sequences of chicken embryo-origin (CEO)vaccine strains, tissue culture-origin (TCO)vaccine strains and some field isolates,which had been published in GenBank. The results showed that: all of 22 isolates in this study, a recent Chinese isolate LJS09 and the CEO vaccine strains were characterized with a 12 nucleotides deletion corresponding to 272 to 283 nt in ICP4 genes,which were different from TCO vaccine strains and WG strain isolated 20 years ago in China. When the phylogenetic tree was constructed,all of 22 isolates were branched together with CEO vaccine strains, but separated from TCO vaccine strains and WG strain. Taken together, the results revealed that there was a close phylogenetic relationship between ILTV field isolates and CEO vaccine strains.
