摘 要:随着能源危机的加剧,纤维素乙醇已经成为可再生能源的重要组成部分。为提高竹质纤维物料的酶水解效率,进行酶解工艺参数优化实验。以预处理丛生竹蒸汽爆破渣为原料,对影响纤维素酶解的4个主要影响因素(包括酶解温度、p H、酶的用量、β-葡萄糖苷酶/滤纸酶比)进行实验,掌握各因素对竹子酶解的最适条件。在此基础上,设计4因素3水平正交优化实验,得出酶解竹纤维的最适工艺条件为:酶解温度为50℃,初始p H 5.2,底物浓度为10%,吐温-20为0.3%,加入的酶量为35 IU/g,β-葡萄糖苷酶酶活/滤纸酶比为1.0,酶解时间为48 h。优化条件确定后,利用2.5 L发酵罐进行放大验证实验,可产生30.37 g/L的还原糖,糖化率为57.49%。本实验所建立的优化条件有利于促进酶与纤维素的催化水解,为后续竹子糖化发酵过程提供参考。As the energy crisis intensifies worldwide, the development of cellulose has become an importantmethod to solve the energy problem. This study is to improve the enzymatic hydrolysis efficiency of bamboofiber material for technical parameters optimization. With sympodial bamboo after steam explosionpretreatment as raw material, a comprehensive investigation of 4 main factors, including temperature, p H, theamount of enzyme, beta- glucosidase/filter paper which affected the enzymatic hydrolysis of cellulose, wasconducted to grasp bamboo enzymolysis optimizing conditions of various factors. An orthogonal optimizationtest was conducted with the four factors to achieve the goal of integration and optimization of enzymatichydrolysis process of bamboo bioethanol. By analysis of the optimization test result, the optimum condition forenzymatic hydrolysis of bamboo was obtained as follows: reaction temperature was 50℃, initial p H was 5.2,substrate concentration was 10%, Tween-20 was 0.3%, the amount of enzyme was 35 IU/g, beta-glucosidaseenzyme activity/ filter paper ratio was 1.0, hydrolysis time was 48 h. After fermentation condition was determined, fermentation amplified verification experiment was performed in 2.5 fermentor. 30.37 g/L ofreducing sugars was generated, and the saccharification rate was 57.49%. The results suggest that the optimumcondition established is useful and would benefit optimizing enzymatic catalytic hydrolysis and can providescientific basis for bamboo fermentation process in the future.
