骨髓间充质干细胞对大鼠胃溃疡影响的实验研究

摘　　要：目的探讨骨髓间充质干细胞对胃溃疡愈合质量的影响。方法大鼠乙酸胃溃疡模型复制3 d后随机分为3组。模型组:只做剖腹手术;磷酸盐缓冲液组:剖腹手术后,在溃疡周围胃壁浆膜下注入磷酸盐缓冲液;干细胞组:剖腹手术后,在溃疡周围胃壁浆膜下注入干细胞。25 d后用组织学方法检测再生胃黏膜扩张腺体数和胃黏液,用免疫组织化学方法检测环加氧酶-2的表达,用酶联免疫测定方法检测胃黏膜前列腺素E2含量,用血流仪检测再生黏膜部位血流量。结果干细胞组的扩张腺体数低于模型组(P<0.01)和磷酸盐缓冲液组(P<0.01),干细胞组的环加氧酶-2和胃黏液积分光密度高于模型组(P<0.01)和磷酸盐缓冲液组(P<0.01),干细胞组的胃黏膜前列腺素E2含量和黏膜血流量高于模型组(P<0.01)和磷酸盐缓冲液组(P<0.01)。结论骨髓间充质干细胞植入溃疡周围能够提高胃溃疡愈合质量。Objective The aim of this study was to explore the effect of bone marrow mesenchymal stem cells （BMSCs） on the quality of gastric ulcer healing. Methods The gastric ulcers were induced with acetic acid in rats. And then the rats with the gastric ulcer were randomly divided into three groups after three days. For the model group, only laparotomy was performed. For the phosphate buffered saline （PBS） group, laparotomy was performed, and then PBS was injected into the serosa around the ulcer. For the stem cell group, laparotomy was performed, and then BMSCs was injected into the serosa around the ulcer. The number of dilated glands and gastric mucus in the regenerative mucosa were detected by histological method. The expression of cyclooxygenase-2 （COX-2） was detected by immunohistochemical method. The content of prostaglandin E2 （PGE2） was detected by enzyme linked immunosorbent assay （ELISA） method. The volume of blood flow in the regenerate gastric mucosa was detected by blood flow instrument after 25 days. Results The number of dilated glands in stem cell group was decreased compared with that of model （P〈0.05） or phosphate buffered saline group （P〈0.05）. The integral light density of COX-2 and gastric mucus in stem cell group were increased compared with that of model （P〈0.01） or phosphate buffered saline group （P〈0.01）. The content of PGE2 and the volume of blood flow in stem cell group were increased compared with that of model （P〈0.01） or phosphate buffered saline group （P〈0.01）. Conclusion The BMSCs transplantation around the ulcer could improve quality of gastric ulcer healing.
