摘 要:目的探讨普萘洛尔和酚妥拉明在神经源性肺水肿(NPE)大鼠中的作用。方法将120只健康雄性Wistar大鼠按随机数字表法分为正常对照组(A组),NPE组(B组),普萘洛尔治疗组(c组),酚妥拉明治疗组(D组),各30只。每组又分为造模后0.5h、6.0h、24.0h3个时间点。除A组外,其余各组采用Marmarous闭合性颅脑损伤模型致大鼠重度、弥散性颅脑损伤。在造模后相应时间点处死大鼠,检测各组肺组织含水量变化,HE染色观察肺组织病理学改变,酶联免疫吸附法检测血清及支气管肺泡灌洗液(BALF)中神经肽Y(NPY)及P物质(sP)水平,免疫组织化学法检测肺组织NPY、sP蛋白表达情况,并进行免疫组织化学评分(IHS)。结果B组与A组相比,伤后24.0h肺组织含水量增高(P〈0.05),伤后6.0h血清NPY水平增高(P〈0.05),伤后6.0h、24.0hBALF中NPY水平增高(P均〈0.05),伤后0.5h、6.0h血清sP水平增高(均P〈0.05),伤后0.5hBALF中SP水平增高(P〈0.05),伤后0.5h.6.0h、24.0h肺组织NPY蛋白表达增多(P均〈0.05),伤后0.5h肺组织sP蛋白表达增多(P〈0.05)。C组与B组相比,伤后6.0h、24.0h肺组织含水量增高(P均〈0.05);伤后6.0h、24.0h血清NPY水平增高(P均〈0.05),伤后0.5h、6.0h及24.0hBALF中NPY水平增高(P均〈0.05),伤后0.5h血清及BALFSP水平增高(P均〈0.05),伤后6.0h肺组织NPY蛋白表达增多(P〈0.05),伤后0.5h、6.0h、24.0h肺组织sP蛋白表达增多(P均〈0.05)。D组与B组相比,伤后6.0h血清NPY水平降低(P〈0.05),伤后6.0h、24.0hBALF中NPY水平降低(P均〈0.05),伤后0.5h血清sP水平降低(P〈0.05),伤后6.0h、24.0h肺组织NPY表达增多(P均〈0.05),伤后0.5h肺组织sP蛋白表达增多(P〈0.05)。结论酚妥拉明能有效抑�Objective To investigate the effecof Propranolol and Phentolamine on neurogenipulmonary edema(NPE) in rats. MethodOne hundred and twenty male Wistaratwere randomly divided into 4 groups:the control group (group A) , the NPE group (group ) , the Propranolol treatmengroup (group ) and the Phentolamine treatmengroup (group D) ,30 casein each group. Diffuse brain injury wainduced in the latte3 groups. The lung wet/dry ratio wacalculated. HE staining waused to measure the histological changein the lung tissues. The levelof neuropeptide Y (NPY) and substance P (SP) in the serum and the bronchoalveolalavage fluid(BALF) were detected by enzyme - linked immunosorbenassay ( ELISA). The expressionof NPY and SP in the lung tissuewere demonstrated by immunohistochemical staining, and immunohistochemical score(HIS) were measured aftescarifying the animaladifferentime point(0.5,6.0 and 24.0 h afteinjury). ResultCompared with group A, watevolume in the lungin group increased a24.0 h ( P 〈 0. 05 ) ; NPY contenin the serum waelevated a6.0 h, while thain BALF waelevated a6.0 h and 24.0 h (all P 〈 0.05) ; SP contenin the serum waelevated a0.5 h and 6.0 h (all P 〈 0.05), while thain BALF waelevated a0.5 h ( P 〈 0.05 ) ; The expression of NPY protein in the lung tissue increased a0.5 h, 6.0 h and 24.0 h ( all P 〈 0.05 ), while the level of SP protein increased a0.5 h ( P 〈 0. 05 ).
