摘  要:[目的]为建立拟除虫菊酯类农药多残留免疫分析方法,对拟除虫菊酯通用人工抗原的合成鉴定进行研究。[方法]根据拟除虫菊酯农药公共结构设计合成了3种通用半抗原,并通过高分辨质谱和核磁共振氢谱对其结构进行鉴定。通过活性酯法将3种通用半抗原分别与多聚赖氨酸(PLL)和牛血清蛋白(BSA)偶联制备成免疫抗原和包被抗原,采用紫外吸收法鉴定。将3种免疫抗原免疫小鼠,制备了拟除虫菊酯抗血清,非竞争间接酶联免疫吸附分析方法测定抗血清效价。[结果]通用半抗原和人工抗原制备成功,合成的人工抗原具有较高的免疫原性,3种抗血清效价分别达到3.2×104、3.2×104、6.4×104。[结论]成功制备了拟除虫菊酯通用人工抗原,为拟除虫菊酯农药多残留免疫分析的方法研究奠定了基础。[Aims] This study was to develop a multi-residue immunoassay for pyrethroid insecticides. The artificial antigens ofpyrethroid insecticides were synthesized and identified in the study. [Methods] Three generic haptens were designed and synthesized based on common structures of pyrethroid insecticides. Their structures were confirmed by 1H NMR and HRMS. The general haptens were conjugated with carrier proteins PLL by activated ester method to preparationimmunogens. This method was also used to conjugate haptens with BSA to preparation coating antigen. [Results] Using UV absorption spectrometry was applied usefullt to determine the conjugations. The mouse immunized with the immunogens and titers of three antiserums (3.2×10^4, 3.2×10^4, 6.4×10^4) were determined by non-competitive indirectenzyme-linked assay procedure. [Conclusions] These results showed that these artificial antigens can be used to study an enzyme-linked imrnunosorbent assay forpyrethroid insecticides.
