摘 要:NK细胞是机体天然免疫的主要承担者,无需肿瘤特异性抗原识别便可以直接杀伤肿瘤细胞,是肿瘤免疫治疗的重要效应细胞,在机体早期抗肿瘤和免疫监视中具有重要作用。近年来,NK细胞的过继免疫治疗获得了长足的进步引起了人们的重视,但由于NK细胞在体内数量较少,制约了其大规模研究试验。因此,研制高效的体外扩增技术是NK细胞过继免疫治疗研究进展的关键。目前,NK细胞的体外扩增培养中除选用合适的细胞来源和培养基外,还需要细胞因子、饲养细胞及其他因素的刺激作用。文章结合近年来文献报道就NK细胞体外扩增技术的影响因素作了综述,以期对已有扩增技术的进一步优化提供有价值的参考。NK cells,as the main undertaker of innate immunity,are equipped to kill tumor cells directly without tumor-specific antigen recognition. What's more,NK cells are the essential effectors for immunotherapy aganist cancer which play an important role in the early organism defense against tumor and immune surveillance. In recent years,the considerable development of NK cells' adoptive immunotherapy has aroused people's attention. However,the number of NK cells in vivo is small which constrains its large-scale experiments and research,so preparing efficient proliferation in vitro is the crucial procedure for studying NK cells' clinical adoptive immunotherapy. Currently,NK cells are nurtured in vitro by cytokines,feeder cells and other stimulating factors besides the appropriate choices of cell sources and media. In this paper,the research progress of proliferation in vitro of NK cells reported recently is reviewed so as to provide valuable reference for further optimizing the existing amplification schemes.
