摘 要:目的观察肝脏保护剂(海迪克胶囊)对肼类火箭推进剂,单推-Ⅲ(DT-Ⅲ),所致小鼠肝脏损伤的干预作用;为其开发与应用提供基础数据。方法将35只昆明种小鼠随机分为6组:空白对照组(生理盐水灌胃);四氯化碳(CCl4)阳性对照组(按10 mg/kg腹腔注射0.125%CCl4溶液);DT-Ⅲ单纯染毒组(2.35 mg/kg灌胃);3个肝脏保护剂海迪克胶囊低(200 mg/kg)、中(400 mg/kg)、高(800 mg/kg)剂量组,其染毒方法同DT-Ⅲ单纯染毒组。后4组每天灌胃1次,各组均连续处理4周。对小鼠血清及肝组织匀浆中肝功能指标、炎性因子、脂质过氧化产物含量及机体的抗氧化能力进行检测,并对肝组织进行病理学观察。结果 DT-Ⅲ单纯染毒组的谷丙转氨酶(ALT)、谷草转氨酶(AST)和血清总胆红素(TBil)的含量分别为(48.18±5.93)U/L,(110.99±22.05)U/L和(2.42±0.18)μmol/L,接近CCl4阳性对照组,且均明显高于空白对照组(P<0.05);施予高剂量肝保护剂后,ALT和TBil的含量均明显低于空白对照组(P<0.05)。DT-Ⅲ单纯染毒组的肿瘤坏死因子α(TNF-α)、白介素6(IL-6)、丙二醛(MDA)、脂质过氧化物(LPO)和谷胱甘肽过氧化物酶(GSH-Px)含量也接近CCl4阳性对照组,且均明显高于空白对照组(P<0.05);施予高剂量肝保护剂后,上述指标的含量均明显低于空白对照组(P<0.05)。与空白对照组比较,DT-Ⅲ单纯染毒组和CCl4阳性对照组的肝组织中,呈现多发性灶性坏死;而施予肝保护剂后,肝细胞损伤明显减少。结论 DT-Ⅲ可导致小鼠肝损伤,而肝脏保护剂对DT-Ⅲ所致小鼠肝损伤具有明显的保护作用。Objective To observe the effect of liver protective agent ( Haidike capsule) on hepatic dam- age induced by hydrazine rocket propellant ( DT-Ⅲ) in mice, so as to provide basic data for its development and use. Methods Thirty-five Kunming mice were randomly divided into 6 groups: blank control group; positive control group (0. 125% CCl4,10 mg/kg,intraperitoneal injection) ; DT-Ⅲ alone group(2.35 mg/kg) ; and 3 groups of DT-Ⅲ +low(200 mg/kg) , medium(400 mg/kg), and high(800 mg/kg)dose of Haidike capsule. DT-Ⅲ and Hairl- ike capsule were administrated by intragastric infusion everyday. Four weeks later,liver function indexes, inflamma- tory eytokines, LPO, and antioxidative ability in sera and liver tissues were measured. The change of histopathology in liver was observed. Results The content of ALT,AST,and TBil in DT- Ill alone group was (48.18±5.93)U/L, (110.99±22.05) U/L, and (2.42±0.18) μmol/L respectively;which was higher than that in blank control group ( P〈0.05 ), and close to positive control group ; the content of ALT and TBil in DT-Ⅲ + high dose of Haidike cap- sule group was lower than that in blank control group (P〈0.05). The content of TNF-α, IL-6, MDA, LPO, and GSH-Px in DT-Ⅲ alone group was significantly higher than that in blank control group (P〈0.05), and close to positive control group ;while the content of these indexes in DT-Ⅲ + high dose of Haidike capsule group was lower than that in blank control group(P〈0.05 ). Moreover, Haidike capsule could decrease the liver damage according to histopathological results. Conclusion Liver protective agent (Haidike capsule) has protective effects on DT-Ⅲ induced liver injury in mice.
