摘  要:目的:通过观察大负荷训练期间男子摔跤运动员IFN-γmRNA和IL-4mRNA表达水平的变化,探讨针灸预处理方法防治运动性免疫失衡的可能作用机制。方法:20例摔跤运动员,随机分为对照组(10例)和实验组(10例),在8周大负荷训练期间进行实验,分别在大负荷运动训练前,大负荷运动训练后、调整训练后采取外周静脉血共5次,采用实时荧光定量PCR检测外周血γ-干扰素(IFN-γ)和白介素-4(IL-4)细胞因子mRNA的表达量。结果:摔跤运动员IFN-γmRNA表达变化,对照组W8与同组W2和W5相比明显降低(P<0.05),调整后W9明显高于W8(P<0.01);实验组W9高于同组W2和W5(P<0.05),实验组W8与同期对照组相比明显升高(P<0.05)。IL-4 mRNA表达对照组W4、W5、W8、W9均高于同组W2(P<0.05,P<0.01);实验组W5与同组W2、W4相比明显升高(P<0.01),W8、W9与同组W5相比明显恢复(P<0.05),实验组W4、W8与同期对照组相比有统计学意义(P<0.01、P<0.05)。IFN-γmRNA/IL-4mRNA比值变化,对照组W4、W5、W8均低于同组W2(P<0.05,P<0.01);实验组W5明显低于同组W2(P<0.05),W8、W9与W5相比明显恢复(P<0.05),实验组W8和W9与同期对照组相比有统计学意义(P<0.01)。结论:大负荷训练期间运动员通过针灸预处理方法调节IFN-γ和IL-4细胞因子mRNA的表达水平,进而维持运动员Th1/Th2细胞因子的动态平衡,这可能是针灸延缓或防止运动员免疫机能下降或失衡的机制之一。Objective : To evaluate acupuncture and moxibustion pretreatment method for the changes on IFN - γmRNA and IL - 4mRNA expressions in male wrestlers after heavy load training and to explore the mechanism of the exercise - in- duced immune imbalance. Method: Twenty healthy male wrestlers were randomly divided into control group (10) and ex- perimental group (10) ,taking a heavy load training period for eight weeks. The blood sample was collected before (W2) and after ( W4, W8 ) heavy load training and after adjusting training ( W5, W9 ). The changes of IFN - γmRNA and IL - 4mRNA expressions were evaluated by real - time fluorescent quantitative PCR. Result : The level of IFN - γmRNA ex- pression in control group at W8 compared with that at W2 and W5 significantly decreased ( P 〈 0.05 ) and the exoerimen-tal group's level of IFN - γmRNA expression at W9 compared with that at W2 and W5 significantly decreased ( P 〈 0.05 ). Experimental group compared with control group at W8 had significant difference ( P 〈 0.05 ). The level of IL - 4 mRNA expression in control group at W4, W5, W8 and W9 compared with that at W2 was significantly higher ( P 〈 0.05, P 〈 0.01 ). The experimental group's level of IL -4 mRNA expression at W5 compared with that at W2 and W4 was sig- nificantly higher ( P 〈 0.01 ). Experimental group compared with control group at W4 and W8 had significant difference ( P 〈 0.01, P 〈 0.05 ). IFN - γmRNA/IL - 4mRNA ratio changes in control group at W4, W5 and W8 compared with that at W2 were significantly lower (P 〈0.05 ,P 〈0.01 ) and the experimental group's changes at W5 compared with that at W2 was significantly different ( P 〈 0.05 ). Experimental group compared with control group at W8 and W9 had significant difference (P 〈 0. 01, P 〈 0.05 ). Conclusion: Wrestlers during heavy load training through the acupuncture and moxibus- tion preconditioning adjust IFN -'y and IL -4 cytokine mRNA expression level
