黄芩素对人胆囊癌细胞系SGC996细胞活力及细胞锌指X染色体蛋白表达的干预作用

摘　　要：目的探讨中药黄芩提取物黄芩素对人胆囊癌的生物效应及机制。方法将人胆囊癌细胞系SGC996用黄芩素处理48h,采用MTT法检测治疗后SGC996的细胞活力;通过对透过transwell膜的SGC996细胞进行计数,评估黄芩素对胆囊癌侵袭转移能力的影响;通过流式细胞术检测黄芩素处理后,SGC996细胞的凋亡情况,并用Western blot方法检测细胞锌指X染色体蛋白(ZFX)的表达。结果 40、80、160、320μmol/L黄芩素组对SGC996细胞活力的抑制率与对照组比较,差异有统计学意义[(19.3±3.8)%、(37.9±5.8)%、(61.6±7.8)%、(84.2±10.2)%比0,P<0.05]。40、80、160μmol/L黄芩素组对SGC996细胞的凋亡诱导率与对照组比较,差异有统计学意义[(7.5±1.2)%、(16.3±1.9)%、(31.2±2.8)%比(0.5±0.5)%,P<0.05]。40、80、160μmol/L黄芩素组对SGC996细胞转移抑制率与对照组比较,差异有统计学意义[(25.6±6.6)%、(57.3±7.9)%、(84.1±11.9)%比0,P<0.05]。Western blot结果显示,黄芩素对ZFX有显著的抑制作用。结论黄芩素有良好的抗胆囊癌生物活性,其抗肿瘤效应的机制可能与下调ZFX蛋白表达有关。Objective To explore the effect of baicalein extracted from Scutellaria Baicalensis Georgi on gallnladder cercinoma and the underlying mechanism.Methods SGC996 cells were treated with baicalein for 48 h and then were detected for cell viability by MTT assay.Cancer metastasis was evaluated by transwell assay.The apoptosis of SGC996 cells was measured by flow cytometry.The expression of ZFX protein in baicalein-treating SGC996 cells was detected with western blot.Results The cell viability inhibitory rate of SGC996 cells in 40,80,160,and 320μmol/L baicalein groups were 19.3% ±3.8%,37.9% ±5.8%,61.6% ±7.8%,and 84.2% ±10.2%,with a significant difference as compared with control group（0,P 0.05）.The apoptosis rate of SGC996 cells in control group and 40,80,and 160 μmol/L baicalein groups were 0.5%±0.5%,7.5%±1.2%,16.3%±1.9%,and 31.2%±2.8%（P〈0.05）; the inhibitory rate of metastasis in these groups were 0,25.6%±6.6%,57.3%±7.9%,84.1%±11.9%（P〈0.05）.Western blot analysis showed that the expression of ZFX protein in SGC996 cells was significantly suppressed by baicalein.Conclusion Baicalein showed a significant anti-tumor effect on human gallbladder cancer.The mechanism may be due to the down-regulation of ZFX.
