摘  要:目的观察生脉饮、失笑散对阿霉素心脏毒性心衰模型大鼠脂质过氧化的影响。方法 SD大鼠108只分为正常组、模型组、曲美他嗪组、生脉饮高剂量组、生脉饮低剂量组、失笑散组,每组18只。阿霉素腹腔注射制备心脏毒性心衰模型,各药物组予相应药物灌胃(曲美他嗪组、生脉饮低剂量组、失笑散组均为临床等效剂量,生脉饮高剂量组为临床等效剂量4倍),1天1次;正常组予等量生理盐水。灌胃14天后及28天后分别采集标本。酶联免疫法检测血清超氧化物歧化酶(SOD)、丙二醛(MDA)。结果 1灌胃28天,模型组SOD(1.992±0.245)ng/m L,与其余各组比较明显下降(P<0.05);生脉饮低剂量组SOD(4.142±0.362)ng/m L高于失笑散组的(3.643±0.366)ng/m L(P<0.05);生脉饮高剂量组SOD(5.475±0.155)ng/m L,高于低剂量组(P<0.05);2灌胃28天,模型组MDA(4.345±0.170)nmol/m L,与其余各组比较明显升高(P<0.05);生脉饮低剂量组与失笑散组比较无明显差异(P>0.05);生脉饮高剂量组MDA(2.929±0.206)nmol/m L,低于低剂量组的(3.122±0.144)nmol/m L(P<0.05);3各药物组灌胃28天均较灌胃14天SOD提高、MDA明显降低(P<0.05)。结论生脉饮、失笑散均能通过提高心衰模型大鼠血清SOD活性、降低MDA含量,降低阿霉素心脏毒性心衰模型大鼠的脂质过氧化损伤。生脉饮作用优于失笑散。Objective To investigate the effect of Shengmaiyin and Shixiaosan on lipid peroxidation in adriamycin-inducing heart failure rat model.Methods One hundred and eight SD rats were randomly divided into blank control group,model group,trimetazidine group,low-and high-dose of Shengmaiyin groups,and Shixiaosan group,with 18 rats in each group.Heart failure model of rats was induced by peritoneal injection of adriamycin.Every group was given the corresponding drug once per day(trimetazidine,low dose of Shengmaiyin,and Shixiaosan were therapeutic dose and high-dose of Shengmaiyin was 4 times of therapeutic dose); blank control group was given equivalent volume of normal saline.On day 14 and 28 of treatment rats in each group were sacrificed for blood sampling.ELISA was used to detect serum superoxide dismutase(SOD) and malondialdehyde(MDA).Results On day 28 of treatment,the serum SOD in model group(1.992±0.245ng/m L) was significantly lower than that in other groups(P〈0.05); that in low-dose of Shengmaiyin group was higher than that in Shixiaosan group(4.142±0.362ng/m L vs 3.643±0.366ng/m L,P〈0.05); that in high-dose of Shengmaiyin group(5.475±0.155ng/m L) was higher than that in low dose group.On day 28 of treatment,the serum MDA in model group(4.345±0.170nmol/m L) was increased compared with that in other groups(P〈0.05); no significant difference in that was found between low-dose of Shengmaiyin group and Shixiaosan group; but that in high-dose of Shengmaiyin group was lower than that in low dose group(2.929±0.206nmol/m L vs 3.122±0.144nmol/m L,P〈0.05).The serum SOD was increased and MDA was decreased on day 28 compared with those on day 14 in all groups(all P〈0.05).Conclusion Both of Shengmaiyin and Shixiaosan treatment can reduce lipid peroxidation by elevating serum SOD and decreasing MDA in adriamycin-inducing heart failure rats.Shengmaiyin may have better efficacy than Shixiaosan does.
