摘  要:目的研究线粒体电压依赖性阴离子通道1(VDAC1)在多发性骨髓瘤患者中的表达情况及其与骨髓瘤细胞代谢和凋亡的关系。方法选择多发性骨髓瘤患者20例作为观察组,非肿瘤患者10例作为对照组。采用RT-PCR方法检测2组骨髓标本单个核细胞中VDAC1基因的表达情况。结果观察组VDAC1基因表达量明显高于对照组(P<0.05);观察组中Ⅲ期患者VDAC1基因表达量明显高于Ⅰ期及Ⅱ期患者(P均<0.05),Ⅱ期患者高于Ⅰ期患者(P<0.05)。结论多发性骨髓瘤患者VDAC1高表达可能与肿瘤细胞高代谢相关,对肿瘤细胞凋亡起到重要作用。VDAC1将为多发性骨髓瘤的治疗提供新的方向。Objective It is to investigate the expression of voltage-dependent anion channel 1 (VDAC1) in patients with multiple myeloma so as to explore its relationship with tumor cell metabolism and apoptosis. Methods 20 patients with multiple myeloma were selected into observe group and 10 cases of non tumor patients were into control group. The expressions of VDAC1 gene in bone marrow mononuclear cells were detected by RT - PCR in both groups. Results The expression of VDAC1 gene in the observe group was higher than that in the control group (P 〈 0.05). In observe group, the expression of VDAC1 gene of patients in stage Ⅲ was markedly higher than that of patients in stage Ⅰ and Ⅱ (P 〈 0.05 ) , and that in stage Ⅱ was markedly higher than that in stage I (P 〈 0.05). Conclusion The high expression of VDAC1 in patients with muhiple myeloma may he related to the high metabolism of tumor cells, plays an important role in tumor cell apoptosis. VDAC1 will provide a new direction for the treatment of multiple myeloma.
