活血祛瘀方对兔腹主动脉内膜损伤后VEGF的影响

摘　　要：目的探讨活血祛瘀方对家兔血管内皮损伤后血管内皮生长因子(VEGF)表达的影响。方法将健康新西兰大耳白兔40只,随机分为模型组(n=11)、中药组(n=12)、西药组(n=11)、空白组(n=6),模型组、中药组、西药组高酯饲料喂养4周后行腹主动脉内膜剥脱术造模。术后各组予正常饲料喂养,中药组予活血祛瘀方灌胃4周,西药组予舒降之、阿司匹林灌胃4周,模型组、空白组予生理盐水灌胃4周。造模术4周后处死家兔取腹主动脉观察动脉病理形态学改变,用免疫组化法观察各组对血管内皮生长因子(VEGF)表达的影响。结果与模型组比较,中药组抑制VEGF表达。结论活血祛瘀方使兔腹主动脉内膜损伤后对血管内膜增殖早期VEGF有拮抗作用,提示其对血管内皮损伤有一定保护作用,为抗经皮冠状动脉介入治疗术(PCI)术后再狭窄提供实验数据。Objective To investigate the effects of blood-activating and stasis-removing recipe on the activation of intimal hyperplasia expression of rabbits after intimadenudation. Methods Forty healthy New-Zealand White Rabbits were assigned randomly to model group（n = 11）,Chinese medicine group（n = 12）,simvastation group（n = 11）,and blank group（n = 6）. Rabbits in model group,Chinese medicine group and simvastation group received high fat diet feed for 4 weeks before undergoing intimadenudation operations. After the operation, rabbits in all groups received normal diet feed while those in Chinese medicine group were treated with blood-activating stasis-removing recipe for 4 weeks. Similarly,rabbits in simvastation group were treated with simvastation and aspirin for 4 weeks. Model group and the blank group were physiological saline for 4weeks. Rabbits were put to death 4 weeks after mold-making operations and their abdominal aorta were observed for pathological morphological changes. The effects of the expression of vascular endothelial growth factor（VEGF） on artery were observed using immunohistochemical method.Results Compared with model group, Chinese medicine group showed inhibited VEGF expression. Conclusion Blood-activating and stasis-removing therapy strengthens rabbtis＇ antagonism of early stage VEGF after abdominal aorta intima damage and has certain protective effects in cases of vascular endothelial injury. This also provides experimental data for percutaneous transluminal coronary intervention（PCI）.
