摘 要:目的探讨转酪氨酸激酶C(Trk C)基因的神经干细胞(NSCs)移植对脊髓缺血再灌注损伤后热休克蛋白70(HSP70)水平的影响。方法 90只Wistar大鼠,采用阻断降主动脉的方法构建脊髓缺血损伤模型,随机分为3组:(1)单纯阻断组,只进行单纯阻断降主动脉;(2)NSCs移植组,阻断降主动脉术后1 d进行单纯NSCs移植;(3)Trk C-NSCs移植组,阻断降主动脉术后1 d进行转Trk C基因NSCs移植。每组于移植术后24 h、48 h、72 h、1周、2周分别处死3只大鼠取脊髓标本,进行组织病理学检查;并收集尾静脉血进行ELISA检测HSP70浓度。组间比较采用单因素方差分析。结果 90只Wistar大鼠全部存活。术后5个时间点中,3组大鼠脊髓组织细胞形态在术后1周时差异最为明显。组织病理学结果显示,与单纯阻断组和NSCs移植组相比,Trk C-NSCs移植组脊髓组织神经细胞形态最接近正常,凋亡细胞最少。3组大鼠移植术后HSP70水平均升高,于术后72 h达到峰值,Trk C-NSCs移植组各时间点的HSP70水平均高于另外两组,差异有统计学意义(P均<0.01)。结论转Trk C基因NSCs移植对脊髓损伤有保护作用,其机制可能是通过替代受损神经细胞,以及促进HSP70的释放而减轻应激反应。Objective To observe the effects of transplantation of trans-Trk C gene neural stem cells( NSCs) on heat shock protein 70( HSP70) levels. Methods Ninety Wistar rats underwent aortic cross-clamping to build a model of spinal cord ischemia-reperfusion injury. Rats were randomly assigned to three groups,aortic cross-clamping group( Group 1),transplanted NSCs at 1 day after aortic cross-clamping( Group 2),and transplanted trans-Trk C gene NSCs at 1 day after aortic crossclamping( Group 3). At time points of 24 h,48 h,72 h,1 w,and 2 w after operation,three rats of each group were sacrificed to perform histopathological examination. Tail vein blood was collected to detect the expressions of HSP70 by enzyme immunoassay. Groups were compared using One-way ANOVA. Results All 90 rats survived. Among 5 time points,cell morphology of three groups showed significant difference at 1 week after operation. Histopathological results showed that neurons in Group 3were preserved better and showed fewest apoptotic cells compared with the other groups. Levels of HSP70 in three groups increased after operation and peaked at 72 hours. HSP70 in Group 3 weresignificantly higher than other groups at all time points( P〈0. 01). Conclusion Transplantation of trans-Trk C NSCs may protect spinal cord from ischemia-reperfusion injury,possibly through replacing injured neurons and enhancing the synthesis and release of HSP70 which might inhibit stress.
