高血压病血瘀证患者血清对人脐静脉内皮细胞活性和形态的影响

摘　　要：目的:研究高血压病血瘀证患者血清对人脐静脉内皮细胞(HUVEC)细胞活性和形态学的影响,从而为从分子水平探讨保护VEC的作用机制奠定基础。方法:通过高血压病患者血清损伤正常培养的人脐静脉内皮细胞(CRL-1730),分别观察12 h、24 h、36 h各组血清对人脐静脉内皮细胞(CRL-1730)形态学的影响,并通过CCK-8法检测人脐静脉内皮细胞活性,流式细胞术检测人脐静脉内皮细胞CD31的阳性表达率。结果:实验各组与对照组比较,差异有统计学意义(P<0.001);血瘀证组与非血瘀证组比较,差异有统计学意义(P<0.001),血瘀证组与正常组比较,差异有统计学意义(P<0.05)。CD31的阳性率为85.12%。结论:人脐静脉内皮细胞在受到受试者血清中营养物质促进生长的同时,还受到血清中致伤因素的影响,高血压病血瘀证患者血清中的致伤因素与非血瘀证组和正常组比较,可能更多或更强。Objective： To study the serum of patients with hypertension and blood stasis syndrome in human umbilical vein endothelial cells（ HUVEC） affecting cell viability and morphology,thereby laying the foundation for the protection of VEC and investigating the mechanism at the molecular level. Methods： The human hypertensive patients normal cultured human umbilical vein endothelial cell damage in serum（ CRL-1730） were used to observe 12 h,24 h,36 h serum of human umbilical vein endothelial cells（ CRL-1730） affecting morphology and by CCK-8 assay to detect the activity of human umbilical vein endothelial cells and by flow cytometry to detect human umbilical vein endothelial cells CD31 positive rate. Results： The experimental group and control group＇s difference was statistically significant（ P〈0. 001）; blood stasis syndrome group and non-BSS group difference was statistically significant（ P〈0. 001）,and compared with normal blood stasis syndrome group,the difference was statistically significant（ P〈0. 05）. CD31 positive rate was 85. 12%. Conclusion： Human umbilical vein endothelial cells in serum can be promoted to grow by both subjects of nutrients,but also affected by the serum injury factors. Hypertension patients with blood stasis syndrome injury factors may be more or stronger compared with those of non-BSS group and the normal group.
