摘 要:为了解绵羊BMP15基因序列特征和组织表达特征,SNP筛查及其与产羔数性状关联性分析,采用生物信息学方法分析绵羊BMP15基因序列特征;利用qRT-PCR检测BMP15基因在‘湖羊’下丘脑、垂体、心等13个组织中mRNA的分布;通过测序验证了绵羊BMP15基因B2突变位点并与产羔数进行关联性分析.结果表明:绵羊BMP15基因ORF全长1 377bp,编码393个氨基酸残基,包括transforming growth factor-beta(TGF-beta)家族和低复杂性区段(low complexity region).Protfun分析结果显示,绵羊BMP15基因作为激素参与生物学过程的可能性最高.绵羊BMP15基因在公羊十二指肠中表达量最高,显著高于其他组织(P<0.05),而在母羊中,卵巢组织的表达量最高,且显著高于其他组织(P<0.05).B2突变位点于BMP15基因编码区外显子Ⅱ的第718bp处,该突变为C→T突变.绵羊产羔数相关联分析结果显示:该位点与产羔数呈显著相关,其中G+型绵羊产羔数为(2.00±0.78),显著高于++型绵羊产羔数(P<0.05),为(1.38±0.54).绵羊BMP15基因B2突变位点与绵羊产羔数呈显著相关,可用作为提高绵羊产羔数性状的分子标记应用于育种.This study was performed to comprehend the sequence and tissue expression characteristics of BMP15 gene,and the relationship between mRNA expression level of BMP15 gene in different tissues and litter size in sheep.The sequence of BMP15 gene in sheep was obtained from NCBI.The tissue distributions of BMP15 were detected by RT-PCR in 13 tissues such as the heart,hypothalamus and pituitary in sheep.The sheep's BMP15 gene SNP loci were searched by sequencing,and their correlation with the litter size were analyzed.The results showed that the ORF length of BMP15 gene was 1 377 bp,encoding 393 amino acids which included some typical domains such as transforming growth factor-beta(TGF-beta)family and low complexity region.The result of Protfun showed that the possibility of BMP15 gene in sheep actas hormone in biological processes was the highest.RT-PCR assays revealed that BMP15 highly expressed in the hypothalamus tissue in ram(P〈0.05)and highly expressed in the ovary of ewe(P〈0.05).The BMP15 gene polymorphism was associated with the litter size which showed that the litter size of G+ was2±0.773 4,the litter size of++ was 1.380 5±0.541 3,and the allele G could increase 0.62 average litter size.The BMP15 gene was involved as hormone in the process that regulating the sheep breeding,it could be used as a molecular marker for raising sheep litter size.
