摘 要:目的:以雷公藤多苷致卵巢功能损伤建立大鼠模型来研究天癸失序的差异性。方法:分别将3月龄、6月龄、11-12月龄大鼠(各20只)随机分成:正常对照组、雷公藤组,每组各10只。雷公藤组灌服雷公藤多苷75mg·kg-1·d-1,正常对照组灌服同等剂量0.9%氯化钠溶液,均连续30d,日1次。结果:造模后,3月龄和6月龄组卵泡刺激素(FSH)显著上升,雌二醇(E2)、抑制素B(INHB)、抗苗勒管激素(AMH)以及卵巢指数下降,均有显著性差异(P<0.05,P<0.01);11-12月龄正常对照组大鼠较前2个年龄段正常对照组大鼠的FSH水平高,INHB、AMH水平低,差异有统计学意义(P<0.05),与3月龄及6月龄大鼠造模后水平相仿,造模后差异无统计学意义。3个月龄段大鼠造模后均有不同程度的卵巢萎缩,生长卵泡减少,间质纤维化,颗粒细胞层数减少,其中,11-12月龄组大鼠最甚。结论:3月龄及6月龄大鼠卵巢功能损伤程度大致相仿,且有进一步加重的趋势,而11-12月龄初老大鼠在生理上接近卵巢功能衰退期,造模后,卵巢储备功能明显下降。Objective: The differences of Tiangui disorder were researched on rats model induced by tripterygium wilfordii polyglycoside(TWP). Methods: The 3 months aged, 6 months aged, 11 to 12 months aged rats, with 20 rats in each age group, as a total of 60 were randomly divided into normal control group, TW group, 10 rats in each group. TW group was gavaged with 75mg·kg-1·d-1 of TWP, the normal group of three months aged was gavaged with the same dose of 0.9% normal saline, once a day for 30 consecutive days. Results: After modeling, the FSH level was significantly increased in the 3 and 6 months aged groups, while the levels of E2, INHB, AMH and ovarian index were decreased(P〈0.05, P〈0.01); For the normal control groups, the FSH level in the 11 to 12 months aged group was higher than that in the other aged groups, while the INHB and AMH levels lower(P〈0.05), and there was no significant difference after modeling. The model group of each age stage had different degrees of ovary atrophy, decreased the growth of follicles, interstitial fibrosis, decreased layers of granular cells, of which 11 to 12 months aged rats were most obvious. Conclusion: The damage level of ovarian function is similar in the three and six months aged groups with a worse tendency. The ovarian function of 11 to 12 months aged rats is close to a recession in the physical, the ovarian reserve function is decreased obviously after modeling.
