化浊解毒软肝方对非酒精性脂肪肝大鼠TC、TG、MMP-2及TIMP-2的影响

摘　　要：目的:探讨化浊解毒软肝方对非酒精性脂肪肝大鼠血清及肝组织总胆固醇(TC)、甘油三酯(TG)、基质金属蛋白酶-2(MMP-2)及基质金属蛋白酶-2抑制剂(TIMP-2)的影响。方法:选取50只健康SD大鼠,采用高脂饮食制备非酒精脂肪肝大鼠模型,随机分为正常对照组(A组)、模型对照组(B组)、非诺贝特组(C组),化浊解毒软肝方低剂量组(D组)、化浊解毒软肝方高剂量组(E组)5组,监测大鼠血清及肝组织TC、TG、MMP-2、TIMP-2的水平。结果:化浊解毒软肝方能明显降低大鼠血清及肝组织TC、TG、MMP-2、TIMP-2的水平。与C组比较,D组大鼠血清和肝组织TC、TG含量较高(P<0.01),而E组大鼠血清和肝组织TC、TG含量无明显差异,MMP-2、TIMP-2及MMP-2/TIMP-2水平降低更明显(P<0.01)。结论:化浊解毒软肝方对非酒精性脂肪肝大鼠血脂、促肝纤维化因子均具有改善作用。Objective： To investigate the influence of Huazhuo Jiedu Ruangan Formula（HJRF） on total cholesterol（TC）, triglyceride（TG）, matrix metalloproteinase-2（MMP-2） and tissue inhibitor of MMP-2（TIMP-2） in non-alcoholic fatty liver（NAFLD） rats. Methods： Fifty healthy SD rats were selected and fed with high fat diet to establish NAFLD rat models, which were randomly divided into 5 groups： control group（Group A）, model group（Group B）, fenofibrate group（Group C）, low and high dosage of HJRF groups（Group D and Group E）. The levels of TC, TG, MMP-2 and TIMP-2 in serum and liver tissue of rats were detected. Results： HJRF could decrease the levels of TC, TG, MMP-2 and TIMP-2 in serum and liver tissue of rats. Compared with that of the Group C, the levels of TC and TG in serum and liver tissue of group D were higher（P〈0.01）, while the difference was not significant between that of group C and group E. The levels of MMP-2, TIMP-2 and MMP-2/TIMP-2 of Group E were decreased significantly when compared with that of Group C（P〈0.01）. Conclusion： HJRF could improve the blood fat and promotion factors of liver fibrosis in rats with NAFLD.
