摘 要:目的:研究解毒祛瘀滋阴方对系统性红斑狼疮(SLE)小鼠肠道菌群的调节机制。方法:30只SPF级8周龄MRL/lpr雌性小鼠随机分为模型组、醋酸泼尼松(西药)组和解毒祛瘀滋阴方(中药)组,每组10只;另设10只SPF级8周龄C57BL/6J雌性小鼠为正常组。收集各组小鼠粪便样本,提取细菌基因组DNA,用荧光定量PCR定量分析大肠杆菌、肠球菌属、双歧杆菌属、乳酸杆菌属以及拟杆菌属细菌的含量;收集各组小鼠尿液样本进行尿蛋白测定;HE染色观察各组小鼠肾脏组织病理变化。结果:模型组与正常组相比,尿蛋白定量、5种细菌含量均有显著性差异(P<0.01);与模型组相比,西药组和中药组小鼠治疗后24h蛋白尿含量均有所下降(P<0.01),肾脏病理损伤情况也都有所缓解,但西药组小鼠肠道菌群仍然处于失调状态,而中药组小鼠体内5种肠道菌群数量均有显著性变化(P<0.01)。结论:SLE模型小鼠存在较明显的肠道菌群失调,采用解毒祛瘀滋阴方治疗后SLE小鼠体内肠道菌群失衡状态得以修复,趋向于正常组水平,这可能是其治疗SLE作用机制之一。Objective: To observe the regulatory effect of Jieduquyuziyin Prescription on intestinal microbiota of systemic lupus erythematosus(SLE) mice. Methods: Thirty 8-week old SPF grade MRL/lpr female mice were randomly divided into a model group, prednisone acetate(WM) group and Jieduquyuziyin Prescription(CM) group, 10 in each group; another 10 8-week old SPF grade C57BL/6J female mice were set as a normal group. Fecal samples were obtained from the four groups and then bacteria(escherichia coli, enterococcus faecalis, baeteroides, lactobacillus and bifidobacteria) DNA were extracted from those samples, RT-PCR was used to analyze the amount of the bacteria; Urinary samples were collected from the four groups and determined the content of proteinuria; HE staining was used to detect the pathological changes of the kidneys. Results: Compared with the normal group, the contents of urine protein and bacteria had significant difference in the model group(P〈0.01); compared with the model group, the urine protein in the WM and CM groups was decreased(P〈0.01) and the pathological injuries were relieved. The WM group had no prominent regulation effect on intestinal microbiota while the CM group showed prominent regulation effect(P〈0.01). Conclusion: There exists alteration of intestinal microbiota in SLE mice. And the Jieduquyuziyin Prescription has a significant regulation effect on intestinal microbiota, which might be one of the mechanisms of treating SLE.
