摘 要:目的:观察活血通督汤对脊髓缺血再灌注损伤(SCII)脊髓组织肿瘤坏死因子(TNF-α)、白细胞介素(IL)-1β、IL-8表达的影响,探讨其作用与机制。方法:72只新西兰家兔随机分为假手术组(n=8)、模型组(n=32)、活血通督汤组(n=32)。模型组、活血通督汤组采用夹闭腰动脉法制作SCII模型。假手术组不夹闭腰动脉,其它步骤同模型组。采用免疫组化法检测脊髓缺血再灌注后0.5、1、4、8h后脊髓组织中TNF-α的表达情况,ELISA法检测各观测时点IL-1β、IL-8的表达情况。结果:假手术组TNF-α、IL-1β、IL-8表达较低;模型组和活血通督汤组TNF-α、IL-1β、IL-8在0.5h即开始增高,并随再灌注的时间延长而持续增高;与模型组比较,活血通督汤组各时间点TNF-α、IL-1β、IL-8的表达显著减少(P<0.05,P<0.01)。结论:活血通督汤作用机制之一是通过减少脊髓组织的TNF-α、IL-1β、IL-8表达,抑制炎性反应,从而减轻SCII。Objective: To investigate the effects and mechanism of Huoxue Tongdu Decoction(HTD) on the the expression of tumor necrosis factor-α(TNF-α), interleukin(IL)-1β, IL-8 in spinal cord ischemic-reperfusion injury(SCII). Methods: Seventy-two New Zealand rabbits were randomly divided into a sham operation group(n=8), a model group(n=32), and a HTD group(n=32). Lumbar artery occlusion was performed to induce SCII model in the model and HTD groups. The expression of TNF-α was observed by immunohistochemical method and the expressions of IL-1β, IL-8 were observed by ELISA method at the time points of 0.5, 1, 4, 8h following perfusion. Results: The expressions of TNF-α, IL-1β, IL-8 were lowest in the sham operation group at each time point, and continuously increased in the model and HTD groups from the time point of 0.5h with the time extension. The expressions of TNF-α, IL-1β, IL-8 in HTD group were significantly decreased at each time point(P〈0.05, P〈0.01). Conclusion: HTD could relieve SCII by reducing the expression of TNF-α, IL-1β and IL-8 to inhibit the inflammatory reaction.
