益心解毒方对气虚血瘀证心力衰竭大鼠心肌组织中NOX2和NOX4的影响  被引量:12

摘  要:目的:从氧化应激角度探讨益心解毒方(YXJDF)对气虚血瘀证心力衰竭大鼠心肌保护的作用机制。方法:120只SPF级雌性SD大鼠按体质量分为手术组(110只)和假手术组(10只),手术组利用左冠状动脉结扎术制备大鼠心力衰竭模型,将成模后存活大鼠随机分为7组:模型组,YXJDF高、中、低剂量组,中西结合组,福辛普利组和芪苈强心组,每组10只。治疗4周后取梗死边缘区组织进行western blot及RT-PCR法检测NOX4及NOX2的表达。结果:Western blot示与假手术组相比,模型组的NOX2和NOX4的表达均明显升高(P<0.01);与模型组相比,YXJDF中剂量组可显著下调心力衰竭大鼠心肌组织中NOX2(P<0.01)及NOX4(P<0.05)的蛋白表达,较其它各治疗组更有优势;RT-PCR结果显示各治疗组均可明显下调心力衰竭大鼠心肌组织中NOX4的表达(P<0.01,P<0.05),以低剂量组下降最多。结论:YXJDF对气虚血瘀证心力衰竭大鼠的心肌保护作用,可能是通过其对损伤心肌组织内氧化应激的调控实现的。Objective: To explore the mechanism of Yixin Jiedu Formula(YXJDF) on the myocardial protection in heart failure rats with qi deficiency and blood stasis syndrome from oxidative stress. Methods: One hundred and twenty SPF grade female SD rats were divided into an operation group(n=110) and a sham operation group(n=10) according to the body mass. The operation group was prepared by left coronary artery ligation to establish heart failure model. All the successfully modeled and survival rats were then randomly divided into 7 groups: a model group, high, medium and low dose of YXJDF groups, a combination group, a fosinopril group and a Qili Qiangxin(QLQX) group, 10 in each one. Then the myocardial tissues around scars were isolated, and western blot and RT-PCR were conducted to analyze the expression of NOX2 and NOX4. Results: The western blot showed that when compared with the sham operation group, the expression of NOX2 and NOX4 increased dramatically in the model group(P〈0.01). Compared with the model group, the medium dose of YXJDF group could repress the expression of NOX2(P〈0.01) and NOX4(P〈0.05), which was superior to other treatment groups. RT-PCR indicated that each treatment group could decrease the expression of NOX4(P〈0.01, P〈0.05), especially the low dose of YXJDF group. Conclusion: YXJDF has a myocardial protective effect in heart failure rats with qi deficiency and blood stasis syndrome by adjusting oxidative stress in the injured myocardial tissue.
