摘 要:目的:探究影响制川乌成品质量的关键因素,分析其与炮制过程控制的关系。方法:采用药典法炮制川乌,严格控制浸泡和加热两个炮制过程,以内无干心为指标判断浸泡程度,以内无白心、口尝微有麻舌感为指标判断加热程度,采用高效液相色谱法,对不同炮制品的新乌头碱、次乌头碱、乌头碱、苯甲酰新乌头原碱、苯甲酰乌头原碱、苯甲酰次乌头原碱6种生物碱进行检测,利用SPSS 17.0软件分析制川乌成品质量与炮制过程控制的关系。结果:川乌生品双酯型生物碱含量较低时,制品单酯型生物碱含量均低于药典规定下限。煮法炮制导致生物碱成分大量流失,使得煮制品单酯型生物碱成分含量达不到药典要求,蒸法炮制则可减少生物碱成分流失。结论:同一批药材经相同加热方式炮制后得到的制川乌成品质量稳定,药典法制川乌选择蒸法可更好地保留有效成分。Objective: To explore the key factors affecting the finished product quality of Aconiti Radix and analyse the relationship with process control. Methods: Processing Aconiti Radix by pharmacopoeia method. The processes of soaking without dry heart and heating without white heart and a little hemp tongue tastes were strictly controlled. Six kinds of alkaloids of mesaconitine, hypaconitine, aconitine, benzoyl mesaconine, benzoyl aconine, benzoyl hypaconine in different processed products were detected by HPLC. The relation between the finished product quality and process control was analysed by SPSS 17.0 software. Results: When the diester alkaloids contents of raw Aconiti Radix was low, the monoester alkaloids contents of Aconiti Radix was lower than the pharmacopoeia limit. Boiling resulted in the loss of alkaloids, lead to the monoester alkaloids contents of aconiti Aconiti Radix cannot reach requirements of the pharmacopoeia, while steaming can reduce the loss. Conclusion: Finished product quality after processing of Aconiti Radix with a batch of materials by the same heating mode is stable. Steaming can better keep effective components when processing with pharmacopoeia method.
