摘 要:目的:探讨丹参、川芎嗪联合对肺纤维化大鼠肺组织表达CTGF和b FGF的影响。方法:90只雄性SD大鼠,随机分为空白对照(NS)组、博来霉素(BLM)组,地塞米松(DXM)组和小、中、大剂量丹参联合川芎嗪干预(分别为C1、C2、C3)组。NS组大鼠气管内灌注0.9%氯化钠溶液,其余各组大鼠气管内一次性灌注BLM造模。NS组、BLM组每日给予0.9%氯化钠溶液,DXM组和C1、C2、C3组分别每日给予DXM或不同剂量丹参联合川芎嗪腹腔注射干预,各组分别于气管内灌注药物后第7、14、28天处死大鼠各5只。肺组织HE染色和Masson染色观察肺组织病理改变;免疫组化检测肺组织CTGF和b FGF的表达量。结果:1与BLM组相比,DXM组大鼠各期肺泡炎性反应及纤维化程度均改善,C1组肺纤维化程度没有明显改善,C2组和C3组第14天时肺泡炎性反应及纤维化程度明显改善,第28天时肺纤维化程度明显改善。2与BLM组比较,DXM组、C2组和C3组各期大鼠肺组织CTGF和b FGF表达量均减少(P<0.05);与DXM组比较,C2和C3组各期大鼠肺组织CTGF和b FGF表达量均减少(P<0.05);与C1组比较,C2组和C3组各期肺组织CTGF和b FGF表达量明显减少(P<0.05);C2组和C3组各期肺组织CTGF和b FGF表达量,差异无统计学意义。结论:一定剂量丹参联合川芎嗪可改善肺纤维化大鼠肺泡炎性反应和纤维化,其作用可能与下调肺组织CTGF和b FGF的表达有关。Objective: To investigate the effects of Salvia miltiorrhiza Bge.(Danshen) and ligustrazine on the expression of connective tissue growth factor(CTGF) and basic fibroblast growth factor(b FGF) in lung tissue of rats with pulmonary fibrosis. Methods: Ninety male SD rats were randomized into normal saline(NS) group, bleomycin(BLM) group, dexamethasone(DXM) group and small, medium and large dose of Danshen and ligustrazine(C1, C2 and C3, respectively) groups. The NS group was administrated with 0.9% sodium chloride solution into trachea, while the other groups were administrated with BLM once into trachea to establish model. The NS and BLM groups were treated with 0.9% sodium chloride solution each day, and the other groups were intraperitoneally injected with corresponding drugs. Five rats of every group were sacrificed at day 7, 14 and 28 after administration. HE and Masson staining were performed to observe the pathological changes of lung tissues. The expressions of CTGF and b FGF in lung tissue were detected by immunohistochemical method. Results: 1Compared with the BLM group, the extent of pulmonary alveolous inflammatory reaction and fibrosis in DXM group was improved, while in the C1 group had no difference, in the C2 and C3 groups was improved at day 14 and the extent of fibrosis was improved at day 28. 2Compared with the BLM group, the expressions of CTGF and b FGF in lung tissue in DXM, C2 and C3 groups were lowered at each period(P〈0.05); Compared with the DXM group, the expressions of CTGF and b FGF in lung tissue in C2 and C3 groups were lowered at each period(P〈0.05). Compared with the C1 group, the expressions of CTGF and b FGF in lung tissue in C2 and C3 groups were lowered at each period(P〈0.05); There was no difference in the expression of CTGF and b FGF of lung tissue between the C2 and the C3 group.
