摘 要:[目的]建立白芨液体快速繁殖体系,并通过诱导愈伤组织建立白芨再生体系。[方法]以白芨种子为材料,在不同液体培养基中进行无菌播种,通过调节光照时长和pH来筛选白芨种子萌发的最佳条件。利用组织培养方法对白芨进行愈伤组织诱导,并对愈伤组织进行抗褐化和再分化研究。[结果]白芨最佳液体快速繁殖条件为MS液体培养基,12h光照时长,pH值5.8~6.5;诱导培养基为MS+1.0 mg·L-16-BA+1.5 mg·L-12.4-D,增殖培养基为MS+1.0 mg·L-16-BA+1.5 mg·L-12.4-D+15 mg·L-1抗坏血酸,分化培养基为MS+0.1 mg·L-1 NAA+0.5 mg·L-16-BA,生根培养基为MS+0.8 mg·L-1 NAA+1.0 mg·L-16-BA,有机添加物对白芨有促进作用。[结论]白芨快速繁殖体系能高效快速繁殖白芨,为白芨产业化发展提供技术支持。本研究对白芨的利用和保护具有重要意义,值得进一步研究开发。Objective] This study was attempt to determine rapid propagation methods for Bletilla striata in vitro using liquid medium, then establish the regeneration system for Bletilla striata through callus induction.[Methods] The seeds of Bletilla striata were sowed in different liquid mediums in the sterile environment. The best seed germinating conditions were explored with different illumination time and pH in liquid medium. The callus ,induction from different explants of Bletilla striata with tissue method, were studied in anti-browning and redifferentiation.[Results] The results show that MS medium is the best medium for seeds germination. The best illumination time is 12h·d-1 and the optimal pH is 5.8 to 6.5. MS+1.0 mg·L-1 6-BA+1.5 mg·L-1 2,4-D is the optimal medium for callus induction and MS+1.0 mg·L-1 6-BA+1.5 mg·L-1 2,4-D +15 mg·L-1 ascorbic acid is the best medium for callus proliferation. The optimal medium for redifferentiation is MS+0.1 mg·L-1 NAA+0.5 mg·L-1 6-BA and the optimal medium for rooting is MS+0.8 mg·L-1 NAA+1.0 mg·L-1 6-BA. Adding organics can promote the corm growth. [Conclusion] Rapid propagation system of Bletilla striata, propagating it efficiently and quickly, could provide technical support for the industrialization development of it. This research has a big importance for utilization and conservation of Bletilla striata, which is worthy of further development.
