白豆蔻-白苣胜挥发油提取工艺及抗失眠药效学研究

摘　　要：目的:研究白豆蔻-白苣胜挥发油的提取工艺,观察其对小鼠的镇静催眠作用。方法:采用L9(34)正交表设计实验,优化提取条件,考察药材粉碎粒度、提取时间、液料比对白豆蔻-白苣胜挥发油得率的影响。观察挥发油对小鼠自主活动以及催眠剂量戊巴比妥钠镇静催眠作用的影响。结果:根据极差分析可知,3种因素对白豆蔻-白苣胜挥发油提取率的影响大小为粉碎粒度>提取时间>液料比。挥发油最佳提取工艺为粉碎粒度20目,提取4h,液料比为12倍。白豆蔻-白苣胜挥发油可减少小鼠走动时间及小鼠前肢上抬次数,缩短戊巴比妥所致小鼠入睡的潜伏时间,延长睡眠持续时间。结论:采用水蒸气提取法提取白豆蔻-白苣胜中的挥发油成分简便可行,适合工业化生产,所提取的白豆蔻-白苣胜挥发油具有镇静催眠作用。Objective：To observe the pharmacodynamics for the treatment of insomnia Baidoukou-Baijusheng essential oil and To study the Extraction process of the essential oil from Baidoukou-Baijusheng.Methods：To observe the influence of autonomic activities and sleeping time of baidoukou-baijusheng essential oil at Hypnotic dose of sodium pentobarbital in mice.Using the L9（34）orthogonal experimental design,the extraction conditions were optimized.To investigate the influence of baidoukou-baijusheng essential oil receiving rate of the fineness of pulverization,extraction time and Liquid than.Results：The baidoukou-baijusheng essential oil were decreased the moving time and raise forelimbs,cut downed the latent time at Hypnotic dose of sodium pentobarbital in mice,extended the sleeping time.Based on the range analysis,The influence of three factor of the baidoukou-baijusheng essential oil receiving rate were the fineness of pulverizationextraction timeLiquid than.The Best Group of Extraction process of the baidoukou-baijusheng essential oil was 20 sieve mesh.The extraction time was 4time.The Liquid than was 12 times.Conclusion：The baidoukou-baijusheng essential oil has a sedative and hypnotic effects.The extraction process of the essential oil from Baidoukou-Baijusheng was feasible and simple.Suitable for industrial production.
