化痰除湿法对早期实验性骨关节炎软骨细胞凋亡的影响

摘　　要：目的:探讨化痰除湿法在早期实验性骨关节炎软骨细胞凋亡的应用研究。方法:选用健康Wistar大鼠40只,体质量为(250±10)g,雌雄各半,按随机数字表法将80只大鼠分为A组化痰除湿法组、B组西药阳性对照组、C组模型组、D组空白对照组每组各20只。所有的大鼠均采用改良Hulth造模法进行造模实验,形成骨性关节炎模型,第4周开始给药,干预4周后取大鼠膝关节软骨光镜观察形态学变化,用原位末端标记法(TUNEL法)、免疫组化法分别观察软骨细胞凋亡情况和增殖细胞核抗原(PCNA)表达情况,以及用放免法测定关节液中IL-1、TNFa的含量。结果:A组的软骨细胞凋亡指数小于模型组,而A组的PCNA表达要高于其他各组,化痰除湿法组大鼠关节液中IL-1、TNFa的含量均较其他组显著升高。结论:化痰除湿法在减少大鼠早期膝关节实验性OA软骨细胞的凋亡有着显著作用,可促进软骨细胞的增殖,对早期骨关节炎有较好的治疗作用。Objective： To study the Huatan Chushi Quyu preparation on cartilage cells apoptosis in Experimental with osteoarthritis. Methods： Selected 40 healthy Wistar rats,weighing（ 250 ± 10） g,male and female,according to figures random method 80 rats were divided into Group A Huatan Chusi method in group B as positive control group,group C as a model group,D group was the blank control group in each group 20. All rats were using modified Hulth modeling method modeling experiments,the formation of osteoarthritis model,the first 4 weeks of administration,four weeks after the intervention from rat knee cartilage morphology was observed by light microscopy,in situ end labeling（ TUNEL method）,immunohistochemical staining were observed chondrocyte apoptosis and the expression of proliferating cell nuclear antigen（ PCNA）,and measured by radioimmunoassay synovial fluid IL-1,TNFa levels. Results： Chondrocyte apoptosis index of Hatan Chushi method group is smaller than the model group; while PCNA expression in Hutan Chushi method group than the other groups; synovial fluid of rats in Hutan Chushi method group IL-1,TNFa levels were significantly higher than the other groups. Conclusions： Huatan Chushi method significantly reduces OA chondrocytes apoptosis in Rat knee in early stage,promote the proliferation of chondrocytes,have a better therapeutic effect early osteoarthritis.
