摘 要:目的:观察龙葵总碱对骨髓瘤细胞株RPMI8226细胞凋亡的作用,确定龙葵总碱抑制RPMI8226细胞生长的最适浓度和最适时间。方法:培养RPMI8226细胞,加入龙葵总碱,设置800、400、200、100、50、25、12.5、6.25、3.125mg/L共9个浓度梯度,另设立空白对照组(0mg/L),分别于24、48、72h以MTT法测定龙葵总碱对细胞生长活性的影响。AnnexinⅤ/PI流式细胞分析法检测细胞凋亡,获得细胞凋亡数据后用Mod Fit LT软件进行细胞凋亡相对定量分析。结果:龙葵总碱可抑制RPMI8226细胞株的增殖,其细胞增殖抑制率呈剂量-时间依赖性;25mg/L龙葵总碱处理RPMI8226细胞48h为最适作用浓度和时间。AnnexinⅤ/PI法证实了龙葵总碱可以诱导人多发性骨髓瘤细胞株RPMI8226凋亡。结论:龙葵总碱可抑制人骨髓瘤细胞株RPMI8226细胞增殖,在一定范围内呈时间和剂量依赖性,龙葵总碱可以诱导人多发性骨髓瘤细胞株RPMI8226凋亡。Objective : To observe the effect of the total alkaloids of Solanum nigrum on the apoptosis of myelo- ma cell line RPMI8226, and to figure out the optimal concentration and time for the total alkaloids of Solanum nig- rum to inhibit the growth of RPMI8226 cells. Methods :The cultured RPMI8226 cells were exposed to the total alka- loids of Solanum nigrum at a concentration of 800,400,200,100,50,25,12.5,6.25,3. 125, or 0 mg/L( control group). The effect of the total alkaloids of Solanum nigrum on cell growth was evaluated using the MTT assay at 24, 48, and 72 h. Apoptosis was determined by Annexin V/PI flow cytometry, and the relative quantitative analysis of apoptosis was performed using the ModFit LT software. Results:The total alkaloids of Solanum nigrum inhibited the proliferation of RPMI8226 cells at a dose - and time - dependent inhibition rate. The 25 mg/L and 48 h were the optimal working concentration and time for the total alkaloids of Solarium nigrum to treat RPMI8226 cells. The An- nexin V/PI assay confirmed that the total alkaloids of Solanum nigrum induced apoptosis of human multiple myelo- ma cell line RPMI8226. Conclusion : The total alkaloids of Solanum nigrum inhibit the proliferation of human myelo- ma cell line RPMI8226 at a time - and dose - dependent manner within a certain range. The total alkaloids of Sola- num nigrum also induces the apoptosis of human multiple myeloma cell line RPMI8226.
