摘 要:目的建立检测人胆汁中利奈唑胺浓度的HPLC法。方法以氯霉素为内标,色谱柱为Eclipse XDB C18(250mm×4.6mm,5μm),流动相为甲醇-水(45:55),流速1.0mL·min^1,柱温30%,检测波长254nm。结果利奈唑胺在质量浓度0.50-100mg·L^-1内线性关系良好(r=0.9998),定量下限为0.50mg·L^-1;低、中、高浓度的日间、日内精密度RSD均〈10%,相对回收率分别为98.62%、101.32%、100.64%,提取回收率〉75%。结论本方法操作简便,结果准确,适用于胆汁中利奈唑胺的检测。AIM To establish an HPLC method for the determination of linezolid in human bile. METHODS Chloramphenicol was used as the internal standard. The analytical column was packed with Eclipse XDB-C18(250mm×4.6mm,5μm) the mobile phase was methanol-water(45:55), the flow rate was 1.0 mL·min^-1, the temperature was 30% and the detection wavelength was 254 nm. RESULTS The liner range of linezolid in human bile was 0.50 - 100 mg·L^-1( r = 0.999 8). The limit of quantitation was 0.50 mg·L^-1. The intra-day and inter-day RSD for assaying the bile sample containing three concentration of linezolid were both lower than 10%, the relative recovery was 98.62%, 101.32% and 100.64% ,and the absolute recovery was more than 75%. CONCLUSION The HPLC method to determine linezolid in human bile is simple, high selectivity and sensitivity.
