氯通道阻断剂NPPB对鸡喙外胚间充质细胞增殖分化影响的研究

摘  要:目的研究氯离子通道阻断剂5硝基2(3苯丙胺)苯甲酸(5 nitro 2(3 phenylpropylamino)benzoic acid,NPPB)对鸡喙外胚间充质细胞(chicken mandibular mesenchymal cells,CMMC)增殖及分化能力的影响。方法应用MTT、实时定量PCR的方法,观察NPPB阻断氯离子通道后,鸡喙外胚间充质细胞在AA-BGP诱导成骨过程中,细胞增殖及Sox9 mRNA、Runx2 mRNA及Col10 mRNA的表达的变化。结果在AA-BGP诱导组及无AA-BGP诱导组,NPPB均抑制了CMMC的增殖(P<0.05)。在AA-BGP诱导组及无AA-BGP诱导组,NPPB下调Col10 mRNA及Runx2 mRNA的表达水平均有显著性差异(P<0.05),Sox9 mRNA的表达水平没有显著改变。结论阻断氯离子通道可抑制鸡喙外胚间充质细胞增殖,并影响细胞向肥大化的软骨细胞分化,可能通过调节Runx2水平影响分化。Objective To study the effect of Chloride channel blocker NPPB on the proliferation and differentiation of chicken man- dibular mesenehymal ceils (CMMC). Methods In thestudy,chicken mandibular mesenchymal cells (CMMC) were isolated and ehon- drocyte maturation was induced by aseorbie acid and b-glyeerophosphate ( AA-BGP). The effect of the chloride channel inhibitor NPPB [5-nitro-2-(3-phenylpropylamino) benzoic acid l on regulation of growth,and changes in Sox9, Runx2 and Coll0 mRNA genes expres- sionswere observed with MTT and real-time PCR assays. Results NPPB inhibited the growth of the CMMC with/without AA-BGP treatment(P 〈 0. 05 ). NPPB antagonized expression of typeX collagen and Runx2 (P 〈 0.05 ) with/without AA-BGP treatment,whilst it had no effect on the early chondrocyte differentiation marker Sox9. Conclusions We conclude that chloride channel in CMMC plays an important role in regulating chondrocyte proliferation and plays a major role in the progression of chondrocyte differentiation into hy- pertrophic cartilage. The data from the current study suggest that maybe chloride channel works in ehondroeyte differentiation by regula- ting the Runx2 level.
