摘  要:目的研究N-羟基-N'-苯基辛二酰胺(SAHA)对七氟烷诱发的幼鼠学习记忆障碍的影响并探讨其及治疗机制.方法 6日龄的C57BL/6小鼠每天暴露在七氟烷下2 h,共3 d来模拟临床反复多次麻醉手术,用水迷宫测试来评价学习记忆障碍,用实时PCR检测评价与突触重塑/可塑性相关、调节神经活性或细胞周期的基因的表达.30只幼鼠随机分为2组,每组15只,分别是对照组和七氟烷组,观察七氟烷对幼鼠学习记忆的变化及相关基因表达.60只幼鼠随机分为4组,每组15只,分别是对照组、七氟烷组、SAHA组和七氟烷+SAHA组,观察SAHA对七氟烷诱发的幼鼠学习记忆障碍的影响及相关基因表达.结果 SAHA减轻幼鼠反复多次使用七氟烷后诱发的学习记忆障碍.结论 SAHA能够减轻了七氟烷诱发的新生小鼠学习记忆障碍,其可能成为预防或治疗吸入麻醉药麻醉相关神经毒性的治疗性药物.Objective To study the effect of SAHA on the sevoflurane induced learning and memory impairment in neonatal mice and the mechanisms of it. Methods Six-day-old C57BL/6 mice were exposed to sevoflurane for 2 h daily for 3 days. Morris water maze test was performed to evaluate learning and memory impairments and the expression of genes related to synaptic remodeling/plasticity, or regulated by neuronal activity or the cell cycle were detected by real-time PCR. Thirty mice were randomly divided into 2 groups (n = 15 ). The effect of sevoflurane on the learning and memory impairment in neonatal mice was observed in control group and sevoflurane group respectively. Sixty mice were randomly divided into 4 groups (n= 15 ). The effect of sevoflurane on the learning and memory impairment in neonatal mice was observed in control group, sevoflurane group, SAHA group and sevoflurane+SAHA group respectively. Results SAHA attenuated sevoflurane-induced learning and memory impairments in neonatal mice. Conclusion SAHA could attenuate sevoflurane-induced learning and memory impairments in neonatal mice. Our findings suggest that SAHA may have potential as a therapeutic agent for preventing or treating the neurotoxicity.
