摘 要:目的研究姜黄素对大鼠角膜碱烧伤新生血管的抑制作用和对角膜NADPH氧化酶4(NADPH oxidase 4)表达的影响。方法选用标准化清洁级雄性SD大鼠24只,随机分为4组,每组6只,右眼为实验眼。A组为空白对照组,不做任何处理,B组为碱烧伤模型组,C组为姜黄素治疗组,D组为二甲基亚砜(DMSO)对照组。B、C和D组分别以直径3 mm的圆形滤纸片浸入1 mol/L Na OH溶液中20 s,均匀的贴于大鼠右眼角膜中央40 s,制造大鼠角膜碱烧伤模型,B组结膜下注射生理盐水0.1 ml,C组注射姜黄素溶液0.1 ml,D组注射姜黄素溶剂DMSO0.1 ml,均隔天注射1次,注射7次。第3天、第7天、第10天、第14天裂隙灯下观察并照相记录角膜新生血管(CNV)生长情况,计算CNV面积,数据做统计学分析。第14天处死所有大鼠,摘取右眼球进行石蜡包埋切片,H-E染色观察角膜形态学变化,免疫组化法(S-P)观察角膜NOX4表达,计算机彩色图像分析软件计算平均光密度值,数据做统计学分析。结果 C组新生血管第14天时明显减少退化,角膜透亮,CNV面积和B组比较有统计学差异;H-E染色C组角膜基质层新生血管减少退化;免疫组化A组NOX4仅在角膜上皮层和内皮层微量表达,B组较A组明显表达,C组和B组比较表达下降有统计学差异,D组和B组比较无统计学差异。结论姜黄素可以有效的抑制大鼠角膜碱烧伤新生血管,其效应与抗氧化抑制NOX4表达有关。Objective To study the inhibition of curcumin on corneal neovascularization (CNV)in the corneal al-kaline-burned-rat(ABR)and the effect of curcumin on expression in corneal NADPH oxidase 4. Methods 24 male SD rats were randomly divided into 4 groups with 6 per group and the right eye was observed. A group was blank control group, B group was alkaline-burned negative control group,C group was curcumin treatment group and D group was solvent control group. The rat alkaline burn model was made respectively by pasting the their right eyes for 40 s with a 3 mm round filter paper sheet which has been immersed into 1 mol/ L NaOH solution for 20 s. B groups were subconjunctival injected 0. 9%saline 0. 1ml every other day,C groups were injected 40 uM curcumin 0. 1ml,D groups were injected 5mg/ ml dexametha-sone 0. 1ml and total injections were 7 times. CNV was observed on the 3rd day,7th day,10th day and 14th day under slit-lamp microscope and calculated CNV area. All rats were sacrificed and took full eyes paraffin embedding on the 14th day. The pathological change was observed by H-E staining and the expression of NOX4 in corneal was detected by immunohisto-chemical method. Results CNV of C group decreased obviously on the 14th day. According to the comparison between B group and C group,CNV area was statistically significant. CNV of corneal stromal layer degenerated in H-E staining. In A group,NOX4 was low expression in epithelium and endothelium of cornea. NOX4 of B group was expressed obviously and C group was lower than B group,which had a statistically significant. In addition,there was no significant difference be-tween D group and B group. Conclusion Curcumin is capable of inhibiting CNV effectively in the corneal ABR,and its effect is determined by antioxidant inhibiting the cornea NOX4 expression.
