摘  要:目的观察六味地黄汤对2型糖尿病胰岛素抵抗大鼠环磷酸腺苷(c AMP)及脂肪组织磷酸二酯酶3B(PDE3B)的影响,探讨其改善2型糖尿病的作用机制。方法将80只SD大鼠随机分为空白组和造模组,造模组采用高脂高糖喂养加腹腔注射STZ造模。将成模大鼠分为模型组、罗格列酮组、六味地黄汤组。各给药组每日给予相应药物灌胃,模型组和空白组给予等量生理盐水,连续30 d。常规检测大鼠空腹血糖(FBG),ELISA检测大鼠血清c AMP和胰岛素(INS)水平,Western Blot检测脂肪组织PDE3B蛋白表达,实时荧光定量PCR检测脂肪组织PDE3B m RNA表达。结果与模型组比较,各给药组大鼠FBG、FINS水平降低(P<0.05,P<0.01);六味地黄汤组大鼠脂肪组织PDE3B蛋白及m RNA表达升高、血清c AMP浓度降低(P<0.01)。结论六味地黄汤可上调脂肪组织PDE3B因子表达,降低c AMP浓度,这可能是其改善2型糖尿病大鼠胰岛素抵抗的作用机制之一。Objective To observe the effects of Liuwei Dihuang Decoction on cAMP and PDE3B inadipose tissues of rats with type 2 diabetes;To explore its mechanism. Methods Totally 80 SD ratswere randomly divided into two groups:control group and molding group. Rats in the molding groupestablished the type 2 diabetic models by feeding high sugar and fat diet combined withintraperitoneal injection of Streptozotocin. The successful modeling rats were divided into modelgroup, Rosiglitazone group, and Liuwei Dihuang Decoction group. Administration groups were givenrelevant medicine for gavage, while model group and blank group were given the same amount ofnormal saline for 30 days. FBG levels were detected, and cAMP and INS levels were detected byELISA. The protein expressions of PDE3B in adipose tissues were determined by Western blot. ThemRNA expressions of PDE3B in adipose tissues were detected by RT-PCR. Results Compared withthe model group, the FBG and INS levels in type 2 diabetic rats were reduced in Rosiglitazonegroup and Liuwei Dihuang Decoction group (P 〈0.05, P 〈0.01). In Liuwei Dihuang Decoctiongroup, the protein and mRNA expressions of PDE3B in the adipose tissues were higher, and cAMPlevels were lower than the model group (P 〈0.01). Conclusion Liuwei Dihuang Decoction canincrease PDE3B expression, and decrease cAMP level, which may be one of the mechanisms forimproving insulin resistance in type 2 diabetic rats.
