摘  要:目的研究多穗柯乙醇提取物不同萃取部位及5个主要成分(三叶苷、3'-O-乙酰基根皮苷、根皮苷、根皮素及2'-O-乙酰基根皮苷)改善胰岛素抵抗的活性。方法采用四甲基偶氮唑盐(MTT)法检测多穗柯乙醇提取物不同萃取部位及其5个主要成分对Hep G2细胞增殖的影响;用葡萄糖氧化酶法检测待测样品对Hep G2细胞糖消耗的影响;用高糖(55 mmol·L-1)诱导Hep G2细胞,建立胰岛素抵抗模型,并用葡萄糖氧化酶法检测待测样品对胰岛素抵抗Hep G2细胞糖消耗的影响。结果在有效浓度范围内,多穗柯乙醇提取物不同萃取部位及其5个主要成分对细胞增殖均无显著影响,对Hep G2细胞的糖消耗亦无显著影响;与模型组相比,乙醇提取物、石油醚层、正丁醇层和水层(25及50μg·m L-1)及根皮苷、根皮素(10及50μmol·L-1)和2'-O-乙酰基根皮苷(10及50μmol·L-1)显著升高胰岛素抵抗Hep G2细胞糖消耗。结论多穗柯乙醇提取物不同萃取部位及5个主要成分中,乙醇提取物、石油醚层、正丁醇层、水层以及根皮苷、根皮素和2'-O-乙酰基根皮苷均能提高胰岛素抵抗的Hep G2细胞的葡萄糖消耗,改善Hep G2细胞的胰岛素抵抗。Objective To determine the effect of different fractions and 5 main compounds fromLithocarpus litseifolius on improving Hep G2 cells insulin resistance. Methods The proliferation of Hep G2 cel ls was detected by 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenylterazolium bromide(MTT) assay. The effect of Lithocarpus litseifoliuson the glucose consumption of Hep G2 cells was investigated; insulin resistance cell model was induced by high concentration of glucose(55 mmol·L^- 1) and the hypoglycemic effect of Lithocarpus litseifolius in Hep G2 cells was detected. Results Different fractions and 5 main com pounds fromLithocarpus litseifolius had little impact on the cell proliferation within the range of effective concentration. There were no obvious influences on the glucose consumption of Hep G2 cells. The prevention effects of total extract(25, 50 μg·mL^- 1), petroleum ether fraction, n-butyl-alcohol fraction and water fraction(25, 50 μg·mL^- 1) were remarkable, all of which increased the glucose consumption of insulin resistance Hep G2 cells by at least 50% compared with the model group. Phlorizin, phloretin(10, 50 μmol·L^-1) and 2'-O-acetylphloridzin(10, 50 μmol·L^- 1) had significant effect on the glucose consumption of insulin resistance Hep G2 cells. Conclusion Different fractions and 5 main compounds fromLithocarpus litseifolius, total extract, petroleum ether fraction, n-butyl-alcohol fraction, water fraction, phlorizin, phloretin and 2'-O-acetylph loridzin can significantly promote the glucose consumption of insulin resistant Hep G2 cells and improve the insulin resistance of Hep G2 cells.
