摘 要:茎柔鱼广泛分布在东太平洋海域,分布海域广、群体结构复杂,赤道附近海域(120°W^110°W)群体与秘鲁外海(5°S^20°S)群体是否存在关联,目前尚不清楚。本文通过基因组高通量测序技术获得了12个多态性SSR位点,利用这12个位点,对茎柔鱼赤道海域群体与秘鲁外海群体进行遗传变异分析。结果表明,所有的SSR位点均为高度多态性位点(PIC为0.795~0.945),位点DG07与DG09极显著偏离Hardy-Weinberg平衡(P<0.01)。赤道海域群体(Ho=0.725,Hc=0.897)与秘鲁外海群体(Ho=0.697;He=0.874)均显示出较高的遗传多样性水平,群体间存在显著的遗传分化(Fst=0.020 46,P<0.01)。太平洋赤道海域近表层存在的复杂海流系统有可能阻碍群体间发生基因交流。建议将茎柔鱼赤道海域群体与秘鲁外海群体看作2个管理单元。Dosidicus gigas is an abundant squid species supporting one of the most important fisheries in the Eastern Pacific Ocean.It is extremely urgent to develop more sensitive DNA markers which show high levels of intraspecific polymorphism thus facilitate determination of its genetic structure.Firstly,D.gigas genome was sequenced by Illumina Solex sequencing platform in order to obtain much more polymorphic microsatellite loci.Secondly,polymorphic microsatellite loci were isolated and screened,and 12 of them were then used to analyze the population genetic variation of D.gigas from equatorial waters(120°W^110°W)and off the Peruvian Exclusive Economic Zones(EEZ,5°S^20°S).Results showed that all the 12 loci were highly polymorphic as polymorphic information content(PIC)varied between 0.795 and 0.945,and loci DG07 and DG09significantly deviated from Hardy-Weinberg equilibrium after Bonferroni correction(P〈0.01).Populations from equatorial waters and off the Peruvian EEZ showed high genetic diversities.Their Howas 0.725 and 0.697,respectively;while their Hewas 0.897 and 0.874,respectively.The pairwise fixation index F-statistics(Fst)was 0.020 46,low but extremely significant,which indicated that significant genetic differentiation existed between two populations.Such differentiation may be due to the complicated current system in upper layer of equatorial waters.We suggested that D.gigasfrom equatorial waters and off the Peruvian EEZ should be considered as two management units.
