摘 要:目的确定1株快速增殖HAV临床分离株HAV302的基因型及其特点。方法提取培养病毒RNA模板,通过反转录-聚合酶链反应技术扩增该病毒编码区基因并行序列测定,随后利用序列分析软件对其进行序列分析和基因型比对。结果 HAV302株VP1/2A区基因序列与Gen Bank中收录的HAVⅠ型中的ⅠB亚型MBB株核苷酸同源性最高(99.4%)。进化树分析显示,HAV302株与MBB株亲缘最近。与MBB株相比,非结构蛋白编码区中的2B和2C区在整个病毒的编码区中变异最大。结论 HAV302株属于HAVⅠ型中的ⅠB亚型,其2B和2C区的变异可能是导致病毒快速增殖的原因之一。Objective To determine the genotype and its characteristics of HAV302, a clinical isolated strain of fast duplicated HAV. Methods The RNA template of HAV302 was extracted, and the gene of the coding region of HAV302 was amplified by RT-PCR and sequenced. The sequence analysis and genotype comparison were carried out by DNA STAR sequence analysis software. Results The gene sequence of the VP1/2A region of HAV302 strain demonstrated a highest homology(99.4%) with the nucleotide sequence of MBB strain of HAV subtype Ⅰ B from Gen Bank. The phylogenetic tree analysis showed that HAV302 strain had the nearest genetic relationship with MBB strain. Compared with MBB strain, the 2B and 2C region of the non-structural protein coding region of HAV302 had the greatest mutation. Conclusions HAV302 belongs to subtypeⅠ B of HAVⅠ. The genovariation of its 2B and 2C region may be one of the reasons for fast duplication of HAV.
