视黄醇结合蛋白4在2型糖尿病组和血糖正常组胆囊组织中的表达差异

摘　　要：目的探讨视黄醇结合蛋白4(RBP4)的蛋白表达量及RBP4 m RNA表达量在胆囊结石合并2型糖尿病组和未合并糖尿病组的胆囊组织中的差异。方法首先应用免疫组化检测30例病例组与30例对照组胆囊组织上RBP4的表达情况,然后用Western blot技术检测胆囊黏膜组织上RBP4的表达量,并与相关临床资料结合进行统计学分析。结果免疫组化检测到RBP4在病例组胆囊组织中的表达强度明显高于对照组(P<0.001),女性表达强度明显高于男性(P<0.001),腹型肥胖者表达强度明显高于非腹型肥胖者(P<0.001);Western blot结果显示:RBP4在病例组的胆囊黏膜组织上的蛋白相对表达量为(2.91±0.86),对照组为(2.17±0.73),差异有统计学意义(P<0.001),病例组RBP4在胆囊黏膜组织中的蛋白表达量明显高于对照组。结论通过免疫组化和Western blot实验,在分子生物学的层面上证明了2型糖尿病组胆囊组织的RBP4表达水平高于对照组,同时女性RBP4的表达强度高于男性,腹型肥胖者RBP4的表达强度高于非腹型肥胖者。[ Objective ] To detect the protein level of retinol binding protein 4 （RBP4） and the expression level of RBP4 mRNA in the gallbladder tissue in gallstone with diabetes and without diabetes. [ Methods ] The expression level of RBP4 in gallbladder tissue of 30 cases in case group and 30 cases in control group was detected using im- munohistochemistry. The relationship between its expression and clinical parameters was analysed. The protein level of RBP4 in gallbladder tissue was further inspected using Western blot. SPSS 17.0 statistical software was used in statistical work. [ Results ] The expression level of RBP4 in gallbladder tissue in case group was significantly higher than in control group by immunohistochemistry experiment （P 〈 0.001）. In addition, the expression level RBP4 was higher in women and the person with abdominal obesity （P 〈 0.001）. The protein level of RBP4 in case group was （2.91＋0.86）, and in control group was （2.17±0.73）, so the protein level of RBP4 in case group was significantly high- er than in control group （P 〈 0.001）. [ Conclusions] The expression level of RBP4 in gallbladder tissue of type 2 diabetes group is justified higher than that of the control group by immunohistochemistry and Western blot experiment, meanwhile, the expression level of RBP4 is higher in women and the person with abdominal obesity.
